增殖性糖尿病视网膜病变患者口服荧光素钠后玻璃体中荧光素钠的含量测定方法的研究

摘    要:目的建立增殖性糖尿病视网膜病变患者口服荧光素钠注射液后玻璃体中荧光素钠含量的高效液相色谱测定方法。方法采用Shim—pack vp—ODS柱(150mm×4.6mm,5μm);流动相:0.1mol·L^-1磷酸二氢钠溶液(pH4.4)-甲醇(1:1);流速:1ml/min;激发波长:470nm、发射波长:513nm。结果人玻璃体中荧光素钠浓度在0.03~0.96μg·ml^-1范围内呈良好的线性关系,r=0.9973。增殖性糖尿病视网膜病变患者口服荧光素钠注射液后玻璃体中荧光素钠含量在0.052~0.375μg·ml^-1范围内。结论方法简便、准确、重现性好,可用于人玻璃体中荧光素钠含量的测定。

Abstract:Aim To establish a high performance liquid chromatography (HPLC) method for determination of fluorescein sodium in vitreous of proliferative diabetic retinopathy patients after taking fluorescein sodium injection orally. Methods Shim-pack vp-ODS was used as the stationary phase, a 0.1mol·L^-1 sodium dihydrogen phosphate (pH4.4)-methanol( 1:1 ) as the mobile phase, with excitation wavelength at 470nm and emission wavelength at 513nm. Results The curve of fluorescein sodium in human vitreous was linear in the range of 0.03 -0.96μg·ml^-1 ( r =0. 997 3). The content of the fluorescein sodium was in the range of 0. 052 -0. 375μg·ml^-1 in vitreous of proliferative diabetic retinopathy patients after taking fluorescein sodium injection orally. Conclusion The method is simple, accurate and reliable and could be used for determination of fluorescein sodium in human vitreous.
