异种(猪)脱细胞真皮基质对大鼠深Ⅱ度烧伤创面肿瘤坏死因子-α、白细胞介素-8、白细胞介素-1表达的影响

摘    要:目的观察异种(猪)脱细胞真皮基质(Xeno-ADM)对大鼠深Ⅱ度烧伤创面中肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)、白细胞介素-1(IL-1)表达的影响。方法取70只6-8周健康雄性Wister大鼠于背部建立深Ⅱ度烧伤模型,并随机分为治疗组和对照组,各组35只。治疗组大鼠烧伤创面采用Xeno-ADM覆盖包扎,对照组大鼠烧伤创面采用传统外用聚维酮碘换药包扎。按分组情况分别于伤后1、3、5、7、10、14、21 d每组各取5个大鼠,沿创缘取创面组织标本,采用免疫组织化学染色法和免疫印迹等技术检测创面愈合过程中炎症介质TNF-a、IL-8、IL-1的表达水平,采用SSPS 11.0统计软件对统计学数据进行处理,组间比较采用配对t检验。结果免疫组化提示:两组IL-8,TNF-a和IL-1均在第1天开始表达,第5天达高峰,第7-21天表达水平逐渐下降。其中,伤后治疗组的TNF-a、IL-8表达均低于对照组,且上述差异均有统计学意义(P〈0.05)。而IL-1的表达在第5天到第7天实验组明显高于对照组,且差异均有统计学意义(P〈0.05)。免疫印记显示,上述检测因子在两组中的表达规律及组间差异均与免疫组化结果相一致。结论 Xeno-ADM可有效抑制创面IL-8、TNF-a的表达和促进IL-1的表达,从而减轻炎症反应,促进创面愈合。

Influence of xenogenic acellular dermal matrix on the expression of Tumor Necrosis Factor-alpha,Interleukin-8, Interleukin-1 in wound healing of deep partial-thickness burned rats

Abstract:Objective To discuss the influence of xenogenic acellular dermal matrix on the expression of Tumor Necrosis Factor-alpha, Interleukin-8, Interleukin-1 in wound healing of deep partial-thickness burned rats. Methods The deep partial-thickness burned models were established in the back of 70 male Wister rats. Rats were randomly divided into experimental group and control group,each group only 35. The rats in experimental group were treated with Xeno-ADM,while the rats in control group were treated by changing the dressing in a traditional way. Samples were collected at 1 d,3 d,5 d,7 d,10 d,14 d and 21 d post-scalding. Immunohistochemistry and Western Blotting were respectively conducted to examine the expression of Tumor Necrosis Factor-alpha, Interleukin-8 and Interleukin-1 in both experimental and control groups. SPSS11. 0 statistical software was used to process the data. Comparision between groups was made using the paired t test. Results Trough Western Blotting,Interleukin-8,Tumor Necrosis Factor-alpha and Interleukin-1 were observed on the 1st day. Their expression reached a peak on the fifth day and gradually declined from the 7th to 21th days. The expression of Interleukin-8 and Tumor Necrosis Factor-alpha in experimental group was lower significantly than that in control group during21 days. The expression level of Interleukin-1 in experimental group was higher significantly than that in control group from the 5th to 7th days. The above differences of the expression between experimental group and control group had statistical significance(P〈0. 05). Immunohistochemistry revealed the expression and differences of IL-1,IL-8 and TNF-a in the both experimental and control groups were consistent with the results of Western Blotting. Conclusions Xeno-ADM can indicate active cells proliferation through downregulating the expression of Interleukin-8 and Tumor Necrosis Factor-alpha and up-regulating the expression of Interleukin-1,accelerating burned wound healing,shorten the treatment courses.
