内皮祖细胞移植对肺动脉高压大鼠一氧化氮、内皮素的影响

摘    要:目的研究同种异体内皮祖细胞(EPCs)移植对野百合碱(monocrotaline,MCT)所致肺动脉高压(Pulmonary Artery Hypertensinn,PAH)大鼠的影响,并观察肺动脉血一氧化氮(NO)、内皮素(ET)的变化,探讨其治疗PAH的机制。方法体外培养并鉴定大鼠EPCs,MCT诱发肺动脉高压模型组由尾静脉注入标记的EPCs。在移植EPCs后第21天,测定肺血流动力学参数,计算平均肺动脉压(mPAP),分别用硝酸还原酶法,放射免疫法测定NO及ET-1值。结果与模型组(29.33±3.01)mm—Hg相比,EPCs治疗组(21.89±2.69)mmHg平均肺动脉压明显下降,肺动脉血的NO值明显增加,(49.28±5.31 vs 21.64±3.06)μmol/L(P〈0.05),ET-1值由(354.40±36.35)pg/ml降至(259.20±29.08)pg/ml(P〈0.05)。结论同种异体EPCs移植可有效降低肺动脉压力,作用机制可能与其分化为血管内皮细胞,修复损伤的肺血管内皮,调节NO与ET-1的含量有关。

Abstract:Objective TO investigate the endothelial progenitor cells transplantation on monocrotaline (MCT) -induced pulmonary hypertension (PAH) rats . Observe the nitric oxide (NO) and endothelin (ET) of pulmonary blood. Disscuss the mechanism. Methods In vitro and identification of EPCs, MCT-induced pulmonary hypertension model group was injected marked EPCs from the tail vein, three weeks after the transplant (21 days), measure pulmonary hemodynamic parameters, calculate the average pulmonary artery pressure ( mPAP), delermine pulmonary blood nitric oxide (NO) and endothelin -1 (ET-1) value with nitrate reduetase and RIA. Results 3 weeks after transplantation, compared with the model, in EPCs treatment group mean pulmonary artery pressure decreased significantly (21.89± 2.69 VS, 29.33 ± 3. OlmmHg), NO value of the pulmonary blood: (49.28 ± 5.31 versus 21.64 ± 3.06 umol /L), ET-1 value of the pulmonary blood : (259.26 ± 29.08 versus 354.40 - 36.35 pg/m]), (P〈0.05). Conclusion EPCs allogeneie transplants can effectively reduce pulmonary artery pressure, the mechanism may be related to vascular endothelial division, repair damage in pulmonary vascular endothelial cells to adjust contents of NO and ET-1.
