TRAF6表达与胃癌患者营养状况及预后的关系

目的 目前有关肿瘤坏死因子受体相关因子6(tumor necrosis receptor-associated factor 6,TRAF6)在胃癌相关领域的研究仍有限.本研究通过检测胃癌患者组织中TRAF6表达,探讨其与患者营养状况和预后的关系,为胃癌治疗提供新思路.方法 采用逆转录-聚合酶链反应、蛋白质印迹法、酶联免疫吸附试验检测2008-01-01-2013-06-30浙江省人民医院102例胃癌患者及53例腹腔良性疾病患者组织(腹直肌、血清)中TRAF6 mRNA和蛋白的表达情况,并结合临床资料分析其对胃癌患者营养状况及预后的影响.结果 胃癌患者腹直肌中TRAF6 mRNA的相对表达量(3.89±0.09)较腹腔良性疾病患者(1.77±1.08)明显升高,t=13.050,P<0.001.蛋白质印迹法结果显示,胃癌患者腹直肌中TRAF6蛋白表达较腹腔良性疾病患者明显升高,P<0.05.胃癌患者血清中TRAF6蛋白的相对表达量(4.89±1.09)较腹腔良性疾病患者(2.78±1.18)明显升高,t=14.595,P<0.001.胃癌患者血清中TRAF6蛋白表达水平与患者体质量丢失(χ2=12.901,P<0.001)、血清白蛋白(χ2=17.422,P<0.001)及血红蛋白水平(χ2=4.886,P=0.027)有关.胃癌患者血清中TRAF6蛋白表达水平与患者胃癌病理类型(χ2=6.154,P=0.046)、淋巴结转移(χ2=10.237,P=0.001)及TNM分期(χ2=7.727,P=0.005)有关.胃癌患者血清中TRAF6蛋白表达水平与患者生存率有关,χ2=6.641,P=0.010.结论 胃癌患者腹直肌及血清中 TRAF6 mRNA 和蛋白表达升高,且血清中 TRAF6 蛋白表达水平患者的营养状况及预后存在关联.TRAF6 可能与胃癌患者癌性恶病质的发生发展有关.

Relationship between the expression of TRAF6 and nutritional status and prognosis in patients with gastric cancer

OBJECTIVE The study aimed to investigate the expression of tumor necrosis factor receptor-associated factor 6 (TRAF6)in patients with gastric cancer and its relationship with the nutritional status and prognosis.METHODS RT-PCR, Western-blot and ELISA were used to detect the expression of TRAF6 Mrna and TRAF6 protein in samples (rectus abdominis muscles,serum)of 102 patients with gastric cancer and 53 patients with benign abdominal diseases.The results were analysed combined with the clinical data.RESULTS The relative expression of TRAF6 Mrna in rectus abdominis muscles of gastric cancer patients (3.89±0.09)was significantly upregulated compared with the control group (1.77±1.08),the difference was statistically significant (t=13.050,P<0.001).The expression of TRAF6 protein in rectus abdominis muscles of gastric cancer patients was upregulated compared with the control group (P<0.05).The relative expression of TRAF6 protein in serum of gas-tric cancer patients (4.89±1.09)was significantly higher than those in the control group (2.78±1.18),the difference was sta-tistically significant (t=14.595,P<0.001).The high expression of TRAF6 protein in serum of gastric cancer patients was sig-nificantly associated with the percentage of weight loss,lower level of serum albumin and lower level of hemoglobin (χ2 values were 12.901,17.422 amd 4.886,P values were <0.001,<0.001 and 0.027).The high expression of TRAF6 protein in serum of gastric cancer patients was related to the pathologic type,lymph node metastasis and TNM stage (χ2 values were 6.154, 10.237 and 7.727,P values were 0.046,0.001 and 0.005).The high expression of TRAF6 protein in serum was related to the survival rate of gastric cancer patients (χ2 =6.641,P =0.010).CONCLUSIONS TRAF6 Mrna and protein are upregu-lated in rectus abdominis muscles and serum of gastric cancer patients.The high expression of TRAF6 protein in serum of gastric cancer patients is related to the poor nutritional status and prognosis,which might play an important role in the de-velopment of cachexia.
