STAT4 rs7574865GG基因型与GT+TT基因型肝癌患者相关危险因素对比分析

目的 一项全基因组关联性研究提出STAT4 rs7574865基因多态性与中国人群乙肝相关肝癌患病风险存在关联.而STAT4 rs7574865基因多态性与环境因素在肝癌发生中是否存在交互作用,鲜见相关报道.本研究探讨旨在STAT4 rs7574865基因多态性与环境因素交互作用对肝癌患病风险的影响.方法 采用单纯病例研究方法,收集2010-01-08-2014-12-23就诊于青岛大学附属医院347例肝癌患者的临床资料和外周血.实时定量PCR检测STAT4rs7574865基因多态性.结果 STAT4 rs7574865基因多态性与乙型肝炎家族史、乙肝e抗体的交互作用经年龄和性别校正后仍存在交互作用.校正后与乙型肝炎家族史相比OR=2.59,95％CI为1.11～6.04;与乙肝e抗体相比OR=1.56,95％CI为1.01～2.41.STAT4rs75748646基因多态性与吸烟史、饮酒史、肝癌家族史、HBV感染、HBV表面抗体、HBVe抗原和HBV核心抗体等无明显交互作用.结论 STAT4 rs7574865基因多态性与乙肝家族史及乙肝e抗体可能存在交互作用,可增加肝癌的发病风险.

Comparative analysis of risk factors of STAT4 rs7574865GG genotype and GT+TT genotype in patients with hepatocellular carcinoma

OBJECTIVE A Genome-wide association study has identified that rs7574865 in STAT4 was associated with hepatitis B virus-related hepatocellular carcinoma(HCC) in Chinese population but the interaction between STAT4 rs7574865 polymorphism and environmental factors with HCC risk remains virtually unknown.We conducted this study to investigate the interaction between STAT4 rs7574865 and environmental factors with the hepatic carcinoma risk.METHODS With a case-only study design,a total of 347 HCC patients' clinical data and blood samples were collected from Jan.2010 to Dec.2014 in Affiliated Hospital of Qingdao University Medical College.The genotype of STAT4 rs7574865 was detected by real-time PCR.RESULTS After adjusted for age and sex,we found that there may be interactions between STAT4 rs7574865 and family history of hepatitis B and HBeAb with the adjusted OR=2.59,95％CI:1.11-6.04 for family history of hepatitis B and OR=1.56,95 ％CI:1.01-2.41 for HBeAb respectively.There was no interaction with other environmental factors including smoking history,drinking history,family history of HCC,HBV infection,HBsAg,HBeAg,HBcAb and STAT4 rs 7574865 with HCC risk in our study.CONCLUSION The interac tions of STAT4 rs7574865 and family history of hepatitis B and HBeAb were found,which may increase the risk of HCC.
