摘    要:目的 探讨茶多酚(Tea Polyphenols,TP)对人宫颈癌HeLa细胞增殖的影响及其机制.方法 MTT比色法检测TP对HeLa细胞增殖的影响;荧光染色及DNA琼脂糖凝胶电泳分析等方法检测TP对HeLa细胞凋亡的影响;Western blot检测Bcl-2、Bax、P53蛋白表达.结果 TP处理HeLa细胞48 h后,细胞生长明显抑制,荧光染色可见明显的核固缩、碎裂现象;DNA凝胶电泳图谱观察到“梯子状”改变;随着药物浓度的增加,Bcl-2蛋白表达量下降,Bax、P53蛋白表达量增加.结论 TP抑制HeLa细胞的生长,诱导其凋亡,其作用机制可能是通过上调P53蛋白,直接激活Bax基因表达,同时抑制Bcl-2蛋白表达来诱导细胞凋亡.

Abstract:Objective To investigate the effect of Tea Polyphenols(TP) on the proliferation of human cervical carcinoma HeLa cells and its possible mechanism. Methods The effect of TP on the growth of cervical carcinoma HeLa cells was tested by MTI assay. Fluorescent staining with Hochest 33258 and agarose gel electrophoresis were used to analyze the apoptosis. The expressions of Bcl2, Bax and P53 were tested by western blotting. Results The inhi- bition rate of HeLa cells elevated after 48h treatment with different concentrations of TP. Nuclei condensation and cleavage can be detected by Fluorescent staining. The formation of typical 'IDNA ladder" can be observed by agarose gel electrophoresis. Western blot showed that TP decreased the expressions of Bcl2 protein and increased the expressions of Bax and P53 protein in a does dependent manner. Conclusions TP can significantly inhibit the proliferation of human cervical carcinoma cells and induce cell apoptosis, Its possible mechanism is likely related to increasing the P53 and Bax levels and decreasing the Bcl2 level
