HCV核心蛋白核酸疫苗转化的减毒鼠伤寒沙门菌在小鼠的免疫原性

摘    要:目的:观察以减毒鼠伤寒沙门菌为载体的丙型肝炎病毒(HCV)口服型核酸疫苗在BALB/C小鼠的免疫原性,探讨用该免疫方法预防丙型肝炎的可行性。方法:将HCV核心蛋白真核表达质粒pcDNA3.1core(核酸疫苗)转化到减毒鼠伤寒沙门菌SL7207,将重组沙门菌和pcDNA3.1core分别口服或肌注接种BALB/C小鼠,检测小鼠的体液和细胞免疫应答及观察其安全性。结果:口服重组沙门菌或肌注接种核酸疫苗均在小鼠诱导相似水平的抗HCV核心蛋白IgG,但口服重组沙门菌诱导的细胞免疫应答明显强于肌注接种核酸疫苗,未见小鼠出现明显毒副反应。结论:减毒沙门菌作为HCV核酸疫苗的载体能增强其诱导的细胞免疫应答,这为研制高效免疫、成本低廉、接种方便的HCV疫苗提供了一个新的可行途径。

Abstract:Objective:To observe the immunogeneicity of an oral hepatitis C virus(HCV) nucleic acid vaccine delivery by attenuated salmonella typhimurium in BALB/C mice and explore the possibility of this immunological method against hepatitis C.Methods:A eukaryotic expression plasmid pcDNA3.1core encoding HCV core protein was transformed into attenuated salmonella typhimurium SL7207,the recombinant bacteria and the plasmid pcDNA3.1core were vaccinated to BALB/C by oral administration or intramuscular injection.The humoral and cellular immune responses and side effects were detected.Results:Two immunization methods could induced IgG against HCV core protein to a similar degree without obvious toxic effect,but oral recombinant salmonella could elicit much stronger cellular immune response than nucleic acid injection did.Conclusion:The cellular immune response induced by HCV uncleic acid vaccine can be largely enhanced by delivery of attenuated salmonella,which maybe indicates a new way for effective,cheap and simple vaccine against hepatitis C virus.
