摘    要:目的 建立分散液相微萃取(DLPME)-高效液相色谱(HPLC)同时测定人血清中维生素A、E含量的方法。方法 2018年5月随机抽取人血液样本50例,经DLPME技术处理后,采用HPLC对人血清中维生素A、E含量进行检测,使用标准曲线法进行定量分析。结果 维生素A标准曲线方程为y=13225x+410.03,相关系数为0.9998,检出限为0.042 μg/ml;维生素E标准曲线方程为y=1383.9x-458.27,相关系数为0.9997,检出限为0.096 μg/ml。维生素A和维生素E的加标回收率分别为90.12%~103.70%和81.09%~92.21%,含量范围分别为(0.62±0.04)μg/ml和(8.41±0.43)μg/ml。结论 DLPME-HPLC法操作简便,节约试剂,对样本富集效率高,通过该方法的研究,对微量血清中维生素A和维生素E的测定具很好的的应用价值。

Abstract:Objective To establish a method for simultaneous determination of vitamin A and E in human serum by dispersive liquid phase microextraction (DLPME)-high performance liquid chromatography (HPLC). Methods In May 2018, 50 human blood samples were randomly selected. After treatment with DLPME technique, the contents of vitamin A and E in human serum were detected by HPLC and quantified by standard curve method.Results The standard curve of vitamin A was y=13225x+410.03, the correlation coefficient was 0.9998, the detection limit was 0.042 μg/ml; the standard curve of vitamin E was y=1383.9x-458.27, the correlation coefficient was 0.9997, and the detection limit was 0.096 μg/ml. The recoveries of vitamin A and vitamin E were 90.12%~103.70% and 81.09%~92.21%, respectively, and the contents ranged from (0.62±0.04)μg/ml and (8.41±0.43)μg/ml.Conclusion DLPME-HPLC method is easy to operate, saves reagents, and has high efficiency of sample enrichment. The research of this method has a good application value for the determination of vitamin A and vitamin E in trace serum.
