洛伐他汀降脂治疗对肾病大鼠血小板源生长因子表达的影响

目的探讨洛伐他汀对阿霉素肾病大鼠血小板源生长因子(PDGF)表达的影响.方法16只Wistar大鼠在阿霉素注射肾病模型后高脂饮食,并随机分为高脂组及治疗组,治疗组给予洛伐他汀4mg/kg,饲养12周,观察血脂、尿蛋白、肾脏病理改变.结果治疗组血胆固醇、甘油三酯、低密度脂蛋白、尿蛋白、肾脏病理积分及泡沫细胞明显低于高脂组,肾脏免疫组化显示治疗组PDGF、纤维连接蛋白减少.结论洛伐他汀可减轻高脂血症造成的肾小球损伤,其机制之一为减少PDGF在肾脏中的表达.

Effects of lovastatin therapy on expression of platelet-derived growth factor in nephritic rats

Objective: To study the effect and mechanism of lovastatin on glomerulosclerosis in adriamycin-induced nephritic rats. Methods: 16 male wistar nephritic rats induced by adriamycin were randomly divided into two groups, the treatment group and hyperlipidemia group. The treatment group was given lovastatin 4 mg/kg. They were fed for 12 weeks. Urinary protein excretion and serum lipid were assayed, and renal glomerulosclerosis index, the expression of platelet-derived growth factor (PDGF) and fibronection (FN) were observed. Results: Serum total cholesterol, triglycerides, low-density lipoprotein were significantly lower in treatment group than that in hyperlipidemia group from the 4 th week on. At the 12 th week, the renal glomerulosclerosis index, the expression of PDGF and FN was also sharply lower in treatment group than that in hyperlipidemia group. Conclusions: Lovastatin could lessen glomerular injury resulting from hyperlipidemia by reducing the expression of PDGF in the renal tissue.
