尿道种子细胞与生物可降解材料的生物相容性研究

摘    要:目的 观察生物可降解材料聚β-羟基丁酸酯(PHB)与种子细胞即兔膀胱移行上皮细胞及平滑肌细胞间的生物相容性,探讨其作为种子细胞载体构建组织工程化尿道的可行性.方法 经过传代培养,分别在PHB与上皮细胞共同培养2、7 d,与平滑肌细胞共同培养1、5 d,每组各取6个样本,用噻唑蓝(MTT)比色法检测细胞的增殖情况,并利用扫描电镜检测细胞在材料上的空间生长情况,评估移行上皮细胞和平滑肌细胞与聚β-羟基丁酸酯的生物相容性.结果 平滑肌细胞能够在该材料上正常生长,各组间A值差异无统计学意义(P>0.05);而移行上皮细胞在该材料上只有少量生长,且实验组与对照组间A值差异有统计学意义(P<0.05),而A、B两实验组间A值差异无统计学意义(P>0.05).结论 聚β-羟基丁酸酯与平滑肌细胞具有良好的生物相容性,但其对移行上皮细胞的生长具有一定的抑制作用,生物相容性不甚理想.

Biocompatibility of biodegradable material to seed cells of tissue engineered urethra
Abstract:Objective To observe the biocompafibility of polyhydroxybutyrate (PHB) to bladder transitional epithelial cells and smooth muscle cells and research the feasibility of PHB as a carder of seed cells to construct tissue engineered urethra. Methods At 2nd and 7th day the transitional epithelial cells were cultured with PHB in vitro,taking 6 samples. So deal with smooth muscle cells at lard and 5thd. Cell adhesion and cell proliferation were detected with MTr assay. Cells were examined by SEM to assess cell growth and morphology. Finally, evaluate the biocompatibility of PHB combined with bladder transitional epithelial cells and smooth muscle cells. Results Smooth muscle cells could adhere to PHB and grew well on it, there is no significant difference between three groups (P > 0.05). Transitional epithelial cells ad-hered to PHB difficultly, there is significant difference between experimental group and control group (P <0.05), but there is no significant difference between A group and B group of experimental group (P >0.05). Conclusion PHB has a good biocompatibility with bladder smooth muscle cells,while has a poor biocompatibility with bladder transitional epithelial cells.
