Effect of dexmedetomidine hydrochloride on proliferation and apoptosis of endometrial cancer cells by regulating miR-223 |

Abstract: | Objective To investigate the effects and mechanism of dexmedetomidine hydrochloride(Dex)on proliferation and apoptosis of endometrial cancer cell line AN3CA.Methods AN3CA cells were divided into control group,Dex groups of different concentrations(1,2,5μmol/L correspongdingly),miR-223 group,miR-NC group,5μmol/L Dex+anti-miR-223 group,5μmol/L Dex+anti-miR-NC group.MTT was used to detect cell proliferation,flow cytometry was used to detect apoptosis,Western blot was used to detect expression levels of CyclinD1,P21,Bcl-2 and Bax protein,and RT-qPCR was used to detect expression level of miR-223.Results Compared with the control group,AN3CA cell activity,the protein levels of CyclinD1 and Bcl-2 decreased in Dex groups(P<0.05),while the apoptosis rate,the protein levels of P21 and Bax,and miR-223 levels increased(P<0.05).Compared with the miR-NC group,AN3CA cell activity,the protein levels of CyclinD1 and Bcl-2 decreased in the miR-223 group(P<0.05),while the apoptosis rate and the protein levels of P21 and Bax increased(P<0.05).Compared with the 5μmol/L Dex+anti-NC group,AN3CA cell activity and the protein levels of CyclinD1 and Bcl-2 increased in the 5μmol/L Dex+anti-miR-223 group(P<0.05),while the apoptosis rate and the protein levels of P21 and Bax decreased(P<0.05).Conclusion Dex may inhibit the proliferation of endometrial cancer cell AN3CA and promote its apoptosis by up-regulating miR-223. |
