大鼠酸性反流性食管炎模型的建立

摘    要:目的 建立一种酸性大鼠反流性食管炎(RE)模型.方法 80只大鼠随机分为实验A、B组和对照C、D组,每组20只.实验组行幽门结扎加前胃结扎术,对照组行胃造口修补术.A、C组术后7 d处死大鼠,B、D组术后14 d处死.模型建立时及取标本前测定食管下段黏膜pH值(pHLEM).术后观察大鼠存活及食管病理改变.结果 4只术中死亡,A、B、C和D组存活率分别为68.4%(13/19)、47.4%(9/19)、83.3%(15/18)和95.0%(19/20),B、D组存活率差异有统计学意义(P<0.01).实验组模型建立前后pHLEM分别为(5.19±0.33)和(3.51±0.87)(P<0.01).实验组(A+B组)RE发生率为81.8%,对照组未发生RE.A、B组RE发生率、食管炎大体评分和镜下评分差异均无统计学意义(P>0.05).结论 幽门结扎加前胃结扎法可成功建立大鼠酸性RE模型.

Establishment of acidic reflux esophagitis model in rats

Abstract:Objective To investigate the probability for establishment of acidic reflux esophagitis (RE) in rats. Methods Eighty Wistar rats were randomly divided into four groups with 20 rats in each group: study groups A and B, and control groups C and D. RE was induced by ligation of the pylorus ring and the forestomach. The rats in control groups underwent repair of gastrostomy, and those in groups A and C were sacrificed at the 7th day after the procedure, and those in groups B and D at the 14th day after the procedure. The pH values of lower esophageal mucosa (pHLEM) were detected during the procedure and sacrifice time. The survival rate of each group was recorded and the histologic changes of esophagus were examined. Results Four rats died during the procedures. The survival rate in groups A, B, C and D was 68.4% (13/19), 47.4% (9/19), 83.3% (15/18) and 95. 0% (19/20), respectively. There was statistically significant difference in the survival rate between groups B and D (P < 0.01). The pHLEM of the rats in the study group before and after the procedures for establishment of RE model were (5. 19 ±0. 33) and (3. 51 ± 0. 87), respectively (P<0.01). 81.8% of the rats in the study group developed RE, while none in the control group. There was no significant differences in incidence of RE, gross morphological score and histological score between groups A and B (all P>0.05). Conclusion It is feasible to establish a successful rat model of acidic RE by ligation of the pylorus ring and the forestomach.
