摘    要:对蛋白质的提取方法进行优化,以便进行念珠菌蛋白质组研究。为使细胞壁有效破碎,除用玻璃珠涡旋、振荡外,加用玻璃棒研磨,提取的蛋白质经过以TCA/丙酮为基础的沉淀、纯化后进行双向电泳。用此方法提取热带、季也蒙、光滑3种念珠菌获得的蛋白质浓度基本相同;玻璃棒研磨能够提高蛋白质的浓度;蛋白质纯化可去除蛋白溶液中的杂质,双向电泳图谱效果良好,没有划线和拖尾。该方法适用于念珠菌蛋白质提取,所获得的蛋白质能够满足双向电泳分析的要求。

Abstract:The method of protein extraction is optimized to study Candida protein group, Besides glass bead was used to vortex and vibrate, glass rod grind was applied to grind to break cell wall into pieces more effectively. After precipitated and purified with TCA/acetone, the extracted protein was subjected to two-dimensional gel electrophorasis. With this extraction method, the protein concentration involving Monilia guilliermondi. Candida tropicals and Candida glabrata was roughly the same, Glass rod grinding can improve the concentration of protein; protein purification can exclude the foreign material from protein solution and the results of two-dimensional electrophoregram are fine without streaking or tailing, This method is suitable for the protein extraction of Candida, and the extracted protein can meet the requirement of two-dimensional electrophoresis analysis.
