川贝母新资源太白贝母中生物碱类成分含量测定

摘    要: 目的 利用高效液相色谱法测定太白贝母中的生物碱成分。方法 采用Agilent Extend C18柱(4.6 mm×250 mm,5 μm,流动相为甲醇-0.02%三乙胺水溶液,梯度洗脱,流速为1.0 mL·min-1,漂移管温度90 ℃;载气体积流量2.3 L·min-1,增益值2,进样量10 μL;柱温25 ℃。结果 贝母辛在28.75~690.0 μg·mL-1,贝母素乙在27.37~560.6 μg·mL-1,贝母素甲在25.75~527.0 μg·mL-1与峰面积的对数值呈线性关系,平均回收率分别为99.4%(n=6,RSD为1.10%;99.7%(n=6,RSD为3.61%;100.3%(n=6,RSD为2.82%。结论 太白贝母中的生物碱成分与川贝母非常相似,为其作为川贝母来源的合理性奠定了基础。

Abstract:OBJECTIVE To develop a HPLC method for determining the contents of peimisine, imperialine and peiminine in Fritillaria taipaiensis. METHODS The analyses were performed on an Agilent Extend-C18 column(4.6 mm×250 mm, 5 μm. The investigated compounds were separated with gradient mobile phase consisting of methanol-water-triethylamine at the flow of 1.0 mL·min-1. The temperature for the detector drift tube was set at 90 ℃ and the nitrogen flow-rate was 2.3 L·min-1, the gain in ELSD was set at 2 in order to obtain the best sensitivity, and the injection volume was 10 μL. RESULTS The investigated compounds including peimisine, peiminine and peimine had good linearity (r2≥0. 9991 within the tested ranges. The average recoveries were 99.4%,99.7% and 100.3% with RSD≤3.61%. CONCLUSION The developed method can be applied to control the quality of the F. taipaiensis with a good accuracy, sensitivity and repeatability.
