VLDL对人尿酸盐转运蛋白hOAT1基因表达的影响

摘    要:目的观察不同浓度极低密度脂蛋白(VLDL)对肾小管上皮细胞(HK-2 细胞)人尿酸盐转运蛋白(hOAT1)mRNA表达的影响。方法根据培养液中所含VLDL浓度的不同,将HK-2 细胞分为:① 单纯采用DMEM培养基组(对照组);② DMEM培养基+50μmol/L VLDL组(V1组);③ DMEM培养基+400μmol/L VLDL组(V2组)。每组培养6瓶细胞,在不同培养液中分别培养48h。采用实时荧光定量PCR检测HK 2细胞中hOAT1mRNA 的相对表达量(2-ΔΔCt 法)。结果所有标本均能检测到hOAT1的表达,且V1组hOAT1 mRNA表达量为对照组的41.6%,V2组为对照组的34.5%。两组与对照组间差异有统计学意义(P<0.05),但两组间差异无统计学意义(P=0.927)。结论VLDL可下调hOAT1mRNA的表达,可能与脂代谢紊乱易合并高尿酸血症有关。

Abstract:Objective To observe the effect of VLDL in different concentrations on the expression of the human urate transporter(hOAT1) gene in HK-2 cells.Methods HK-2 cells were divided into the control(cultured with DMEM),the V1(cultured with DMEM and 50 umol/L VLDL) and the V2(cultured with DMEM and 400 umol/L VLDL) groups.In each group,6 bottles of cells were cultured in vitro for 48 hours,and then the relative quantity of hOAT1 mRNA in the HK-2 cells was determined by real-time fluorescent quantitative PCR assay.Results hOAT1 mRNA could be detected in all samples,and it was significantly higher in the control group than in the other groups,and in the V1 group it was 41.6% of that in the control group and in the V2 group it was only 34.5% of that in the control group.Conclusions VLDL can down-regulate the expression of the hOAT1gene possibly induced by lipodystrophy easily acting with hyperuricemia.
