大鼠移植性骨肉瘤模型的建立

摘    要:目的:建立大鼠移植性骨肉瘤模型。方法:采用大鼠骨肉瘤细胞系UMR106接种Wistar大鼠后肢股骨远端或胫骨近端的骨髓腔中,成瘤后,进行影像学、病理组织学观察及血清生化检查。结果:本方法致瘤率为86.7%,肺转移率为96.2%。影像学可见骨膜反应,溶骨变化。病理组织学可见骨组织、骨骺板和软骨被肿瘤细胞所侵蚀、破坏,核分裂多见,瘤组织血管丰富,可见坏死、颗粒状钙化、新生类骨质样组织。大鼠血清碱性磷酸酶明显高于对照组(P<0.01)。结论:该方法建立的大鼠骨肉瘤模型,其恶性生物学行为接近临床骨肉瘤患者实际情况,为临床骨肉瘤实施新疗法与基础实验研究提供了良好的动物模型。

Abstract:Objective: To establish the graft osteosarcoma model in rats. Methods: Rat osteosarcoma cell line named UMR106 was injected into rat distal femoral or proximal tibia bone marrow cavity. After the osteosarcoma appeared, radiological and pathological methods were used to detect the tumor. Serum biochemical index was also measured. Results: Ostesarcoma was observed in 86.7% rats and the rate of lung metastasis was 96.2%. Periosteal reaction and osteolysis could be seen in the radiological films. Pathological changes included the erosions of bone tissue, epiphyseal plate, and cartilage destroyed by osteosarcoma cell. The karyokinesis was common and there were plenty of blood vessels, osteonecrosis, calcified granules, and neogenetic osteoid tissue in the tumor. The serum ALP levels were significantly higher than that in control group (P < 0.01). Conclusion: The malignant biological activities in osteosarcoma model were similar to that in osteosarcoma patients. This model is an useful tool for the development of clinical new therapy and basic experimental study.
