局部应用血管内皮生长因子基因对大鼠皮瓣不同部位作用的实验研究

摘    要:目的局部应用血管内皮生长因子基因转染大鼠皮瓣,初步探讨其对皮瓣成活率的影响及对皮瓣不同部分促血管作用效果。方法将30只SD大鼠作为实验模型,随机分为3组,每组10只,制作背部随意皮瓣,分别注入脂质体包裹的PcDNAVEGF165(目的基因组,实验组),PcDNA(空白质粒组,对照组),NS(生理盐水组,对照组)。术后4d断蒂,断蒂后7d处死大鼠观察:(1)各组皮瓣成活率比较。(2)取皮瓣远端活组织标本行常规HE染色,检测平均微血管数目及内径。(3)取皮瓣远端活组织标本行VEGF免疫组化染色观察VEGF表达情况。(4)取目的基因组皮瓣远段,中段,近段成活组织标本常规HE染色,检测平均微皮瓣血管数目。结果目的基因组皮瓣平均成活率明显高于空白质粒组和生理盐水组两对照组,平均微血管数目明显多于两对照组,且为管径较小的新生血管,均具有统计学意义(P〈0.05)。免疫组化染色强度高于两对照组。目的基因组皮瓣远端血管数目多于近端,具有统计学意义(P〈0.05)。结论局部注射VEGF基因后可以增加缺血皮瓣血管新生,提高皮瓣成活率,在皮瓣不同部位促血管增生作用不同。

Topical application of vascular endothelial growth factor gene in different parts of the skin flap in rats

Abstract:Objective To explore the topical application of vascular endothelial growth factor gene to rat's skin and its effects on flap survival and flap vascular of the different parts,right flap vascular role of the different parts.Methods 30 rats were divided into three groups (n=10). After the model of flap was established in the rats,PcDNAVEGF165 was injected in purpose gene group;PcDNA was injected in bland plasmid group and NS was injected in control group.After flap formed and drug used,the pedicel was devised at4 days. 7 days after pedicle division,the rats were killed,and then the flap survival rate was measured. The microvessels were study by histological examination. The expression of VEGF was assessed by imunohistochemical staining.Results The average flap survival rate of the target gene group was significantly higher than the blank plasmid group and saline group and control group. The average number of blood vessels significantly more than two control groups. And smaller in diameter and angiogenesis (P〈0.05). Immunohisto chemical staining was higher than that in two control groups.Conclusion After local injection of VEGF gene can increase the angiogenesis of ischemic skin flap to improve flap survival rate,different parts of the flap to promote the role of different angiogenesis.
