帕金森病大鼠颈动脉体球细胞块移植治疗的实验观察

摘    要:为评估颈动脉体球细胞块移植治疗帕金森病(PD)的疗效。采用立体定位注射6-羟多巴胺(6-OHDA)制备偏侧PD大鼠模型,右侧纹状体内分别移植入自、异体颈动脉体和胚鼠中脑组织块,移植后2、4、8和12周记录阿朴吗啡诱发大鼠的旋转行为,测定移植区谷氨酸(Glu)、甘氨酸(Gly)、天门冬氨酸(Asp)、γ-氨基丁酸(GABA)和多巴胺(DA)的含量变化。结果发现:与对照组比较,PD大鼠阿朴吗啡诱发的旋转行为明显改善达12周之久,DA水平升高显著(P<0.01)且以自体颈动脉体移植组最佳;自体颈动脉体移植组纹状体内Glu和GABA含量显著降低(P<0.01)。结果表明:脑内移植颈动脉体球细胞块可明显改善PD大鼠药物诱发的旋转行为,提高纹状体内DA含量并降低Glu和GABA含量,是一种有效治疗PD的方法。

Abstract:In order to observe the effects of glomus cells aggregate within carotid body grafts in a hemiparkinsonian rat model, rats with unilateral 6 hydroxydopamine lesions of the right dopaminergic neurons of substantia nigra received intrastriatum grafts of homo or hero intrastriatum glomus cells within carotid body and fetal mesencephalic neurons respectively. Apomorphine induced rotation was monitored for 40 min each time at 2nd, 4th, 8th and 12th week after grafting respectively. The striatum was cut for Glu, Gly, Asp, GABA and dopamine contents analyzed by HPLC at the end of the experiments. The results showed that as compared with the control group, apomorphine induced rotational behaviors were significantly reduced for 12 weeks and the dopamine contents were significantly elevated, especially in homo graft group whose contents of Glu and GABA were decreased after grafting ( P <0 01), respectively. These findings demonstrate that intrastriatum grafts of glomus cells aggregate within carotid body in rats with 6 OHDA lesions can reduce apomorphine induced rotational behavior and elevate the dopamine contents, decrease the Glu and GABA contents within striatum, is an effective method for Parkinson disease.
