乙酰葛根素对大鼠局灶性脑缺血再灌注损伤的脑组织及血清NO和NOS的影响

摘    要: 目的观察乙酰葛根素对大鼠急性局灶性脑缺血再灌注损伤的NO保护作用。方法采用大脑中动脉内拴线阻断法(MCAO)造成大鼠局灶性脑缺血再灌注损伤模型,测定脑纽织匀浆和血清中NO含量及NOS水平的变化,并进行病理组织学观察结果脑缺血再灌注24,48 h后,模型组脑组织匀浆中NO,NOS活性及血清中NO水平较对照组均有显著性升高(P<0.01),给药组能降低其水平,且与模型组相比有显著性差异(P<0.05)。结论乙酰葛根素能通过降低NO毒性,发挥脑保护作用,减轻缺血再灌注损伤。

Abstract:OBJECTIVE To investigate the neuroproteelive effect of acetylpuerarm (compound N-2211)and peurarm on focal cerebral is-chemia-reperfusion injury. METHODS The NO level and NOS activity in brain tissue and serum were measured in the rats with reversible middle cerebral artery occlusion( MCAO) without cramectomy. RESULTS The levels of NO and NOS in brain homogenate increased significantly in ischemia-reperfusion group,compared with sham group, and there was the same change happened for the level of NO in serum.NO and NOS levels decreased in acetylpuerarin-treated groups and peurarin-trealed group, compared with ischemia-reperfusion rats. At the same time, the number of living pyramidal cells in CA1 region of hippoeampus increased significantly in acetylpuerarin-treated rats and peurarin-treated rats, compared with ischemia-reperfusion rats. CONCLUSION The results suggested that the effect of acetylpuerarin and peurarin on decreasing NO production played a role on the amelioration of focal brain ischemia-reperfusion injury.
