橙皮苷防治糖尿病视网膜病变药效及网络药理学机制研究

摘    要:目的观察橙皮苷对高糖诱导的视网膜血管内皮细胞的影响,并运用网络药理学及分子对接探讨其分子机制。方法在高糖环境下培养视网膜血管内皮细胞,检测橙皮苷对高糖条件下细胞迁移和试管形成的影响。利用Pubchem、Swisstarget和PharmGKB等数据库检索橙皮苷抗糖尿病视网膜病变潜在作用靶点,通过DAVID数据库进行富集分析,采用Accelrys Discovery Studio Client 2.5将橙皮苷和靶蛋白进行分子对接。结果橙皮苷可减少视网膜内皮细胞的迁移数量,同时降低细胞成管能力。网络药理学结果显示,橙皮苷抗糖尿病视网膜病变靶点共17个,KEGG通路富集得到8条信号通路。结论橙皮苷可以抑制高糖诱导下内皮细胞迁移和管道形成,提示橙皮苷可能具有防治糖尿病视网膜病的药效,其机制可能与影响表皮生长因子受体、血管内皮生长因子-A、雌激素受体1和雌激素受体2等有关。

Abstract:Objective To observe the effect of hesperidin on the retinal vascular endothelial cells induced by high glucose,and to explore its molecular mechanism by the network pharmacology and molecular docking.Methods The retinal vascular endothelial cells were cultured in the high glucose,and the effects of hesperidin on cell migration and tube formation were detected.PubChem,Swisstarget and PharmGKB database were used to search the potential targets of hesperidin and diabetic retinopathy.The target protein and hesperidin were linked by Accelrys Discovery Studio Client 2.5.Results Hesperidin could reduce the migration and tube formation of the retinal endothelial cells.The results of network pharmacology showed that there were 17 targets of hesperidin against diabetic retinopathy,and 8 signal pathways were enriched by KEGG pathway.Molecular docking confirmed that hesperidin combined with epidermal growth factor receptor.Conclusion Hesperidin can inhibit endothelial cell migration and tube formation induced by high glucose,and its mechanism may be related to epidermal growth factor receptor,vascular endothelial growth factor-A,estrogen receptor 1 and estrogen receptor 2.
