摘    要:目的 构建慈丹胶囊活性成分-作用靶点网络和蛋白相互作用网络,对靶点涉及的功能以及通路进行分析,探讨慈丹胶囊抗肝癌作用机制。方法 通过TCMSP数据库(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform, http://lsp.nwu.edu.cn/tcmsp.php)挖掘获取慈丹胶囊中丹参、当归、黄芪、莪术、鸦胆子、马钱子、冰片、牛黄、山慈菇的主要活性成分。利用GeneCard数据库(http://www.genecards.org/)和OMIM数据库 (Online Mendelian Inheritance in Man,http://www.omim.org/)预测和筛选慈丹胶囊活性成分对应靶点中与肝细胞癌相关靶点。采用Cytoscape构建活性成分-作用靶点网络,采用String数据库(http://string-db.org/)和Cytoscape软件绘制蛋白相互作用网络。采用DAVID数据库(The Database for Annotation, Visualization and Integrated Discovery,https://david.ncifcrf.gov/)对靶点进行GO (Gene ontology) 及KEGG(Kyoto Encyclopedia of Genes and Genomes )通路分析。结果 筛选得到慈丹胶囊活性成分24个,共得100个作用靶点。网络分析结果表明,慈丹胶囊主要涉及细胞过程、生物调控、代谢过程、对化学刺激反应等生物过程。靶点通路分析结果显示,慈丹胶囊抗肝癌作用的靶点主要涉及TNF、PI3K-Akt、MAPK等信号通路。结论 慈丹胶囊抗肝癌作用具有多成分、多靶点、多通路的特点,其可能通过调节TNF、PI3K-Akt、MAPK等相关通路发挥作用。

Abstract:Objective This study was designed to explore the mechanism of the effect of Cidan capsule on hepatocellular carcinoma through establishing the active components-targets network and protein interactions network and analyzing the functions and pathways Methods The main active components of Cidan capsule were obtained through TCMSP database and literature study. The targets to treatment hepatocellular carcinoma of the active components of Cidan capsule were predicted and screened by GeneCards and OMIM. Cytoscape software was used to construct the active component-target network and String database and Cytoscape software were used to construct the protein interaction network. GO and KEGG pathways involved in the targets were analyzed using DAVID database. Results The results showed that 24 active components of Cidan capsule were screened and 100 targets were obtained. The results of network analysis showed that cellular processes, biological regulation,metabolic process and response to chemical stimulus were mainly involved in the effect of Cidan capsule on hepatocellular carcinoma. The results of target pathway analysis showed that the targets of Cidan capsule for hepatocellular carcinoma were mainly involved in TNF,PI3K-Akt,MAPK and other signaling pathways. Conclusion The effect of Cidan capsule on hepatocellular carcinoma is characterized by multiple components, multiple targets and multiple pathways, and it may play a role by regulating TNF,PI3K-Akt,MAPK and other related pathways.
