摘    要:目的筛选建立一种快速准确检测肠出血性大肠杆菌O157:H7的方法,初步了解在本地区家禽、家畜养殖场和肉类食品中的带菌状况.方法以自动酶标免疫测试系统(VIDAS)、自动免疫磁珠收集系统(AIMS)与传统常规分离方法进行比较.结果应用自动免疫磁珠收集系统对79份实样的检测,检出率为6.33%,在4天内能做完全鉴定结果.该法能检出<10 cfu/ml模拟样品中的O157:H7,同时选择使用CHROMagar O157琼脂平板的效果要明显优于山梨醇麦康凯(SMAC)琼脂平板.自动酶标免疫测试系统和传统分离方法未检出.结论以自动免疫磁珠收集系统结合CHROMagar琼脂平板建立的O157:H7分离方法具有特异性好、敏感性高、快速简便的特点,可以用于O157:H7外环境的监测和食品污染源调查.

Abstract:Objective To screen and set up a rapid and accurate screening method and to detect the Escherichia coli O157:H7.To find out the obtain carrier rate of Escherichia coli O157:H7 in domestic animals, fowls and meat products.Methods VIDAS and Automated Immunomagnetie Separation System (AIMS) were used to compare with traditional separation method. Results Sev-enty-nine samples were tested with Automated Immunomagnetie Separation System and a complicated result of 6.33% positive rate of Escherichia coli O157:H7 was obtained within 4 days. This technique can identify O157:H7 in simulated samples, which was less than 10 cfu/ml. Meanwhile, the efficiency of using CHROMagar O157 agar plate was better than that of using SMAC plate. Both VIDAS and traditional methods could not detect any O157:H7 when the bacteria concentration was lower than 10 cfu/ml. Conclusion Using the method of combining Automated Immunomagnetic Separation System (AIMS) with CHROMagar to separate O157:H7 gives the characteristics of rapid, easy use, high sensitivity and more specificity. It can be used in surveillance on environment and inspection on the source of food contamination.
