何首乌对记忆和抗疲劳作用的实验研究

摘    要:目的研究何首乌对小鼠学习记忆的影响及其抗疲劳作用。方法采用跳台试验、水迷宫实验与爬杆试验、负重游泳试验和肝糖原测定(抗疲劳作用),分别观察低、中、高剂量(2.6、7.8、23.4g/kg)何首乌提取物对小鼠改善记忆作用和抗疲劳作用的影响。结果①跳台实验结果显示高剂量组在第1天测试和24h后重测试时,动物平均错误次数显著减少(P〈0.05);水迷宫实验结果显示高剂量组动物平均错误次数减少(P〈0.05)。②在抗疲劳实验中,爬杆试验的结果显示中高剂量的何首乌提取物能够显著延长小鼠在有机玻璃杆上的坚持时间(P〈0.05)。负重实验发现高剂量组动物负重游泳的生存时间与正常组相比明显延长,解剖后的肝糖原测定发现低剂量组肝糖原量明显降低。结论 23.4g/kg的何首乌对小鼠的记忆获得有改善作用,用药量大于7.8g/kg的何首乌对抗疲劳功能有明显改善作用。

Abstract:Objective To explore the memory and anti-fatigue effects of Polygonum multiflorum on mice. Methods The effects of Polygonum multiflorum extract on memory and anti-fatigue in mice were studied by step down test,Morris water maze test,pole-climbing test,weight-loaded swimming test and hepatic glycogen test (anti-fatigue experiment) at low,medium and high doses (2.6,7.8,and 23.4 g/kg) ,respectively. Results ①In the step down test,the average number of mistakes reduced significantly in the mice in the group intragastrically administered with Polygonum multiflorum at high dose in the first day of testing and the re-test after 24 hours (P0.05),while fewer error times were recorded in the group of mice intragastrically administered with Polygonum multiflorum at high dose in water maze test;②In anti-fatigue effect experiments,it was showed that the time in the glass rod was prolonged significantly in the group of mice intragastrically administered with Polygonum multiflorum extract at high doses (P0.05). And it was found that the survival time of swimming of the mice fed with Polygonum multiflorum extract at high doses in weight-loaded swimming test was significantly longer than that in the normal group. Significant decrease of liver glycogen in the group ingrastrically administered with Polygonum multiflorum extract at low dose was found in hepatic glycogen assay after biopsy.Conclusion Memory-improving effects in mice intragastrically administered with Polygonum multiflorum extract at the dose of 23.4 g/kg were observed,and anti-fatigue effects in mice intragastrically administered with Polygonum multiflorum extract at the doses higher than 7.8 g/kg were observed.
