丙型肝炎病毒NS3与干扰素调节因子3的相关关系研究

目的了解丙型肝炎病毒(HCV)非结构区蛋白质NS3与干扰素调节因子3(IRF3)的相关关系。方法①建立病毒感染条件下IRF3核内移的实验室检测方法;②构建包含HCV NS3基因片段的质粒;③建立免疫荧光检测方法,分析NS3对IRF3核内移的影响,进而明确二者的关系。结果病毒感染和HCV NS3同时作用可以明显降低IRF3的核内移率。结论HCV NS3具有调节IRF3功能的作用。HCV有可能通过抑制IRF3来实现病毒免疫逃避。

The study on the relationship between interferon regulatory factor 3 and NS3 of HCV

Objective To investigate the relationship between the HCV NS3 protease and interferon regulatory factor 3(IRF-3).Methods In the present study,first set up the experiment for the detection of IRF,then the plasmid containing HCV NS3 genome was constructed.Finally the relationship between the NS3 and endogenous IRF-3 was studied.Results IFA results showed that comparing with activating effect of virus infection alone,collective action of transfection and virus infection could more effectively reduce the nuclear translocation of endogenous IRF-3 of HeLa cell.Conclusion HCV NS3 protease displayed obvious inhibition of nuclear translocation of endogenous IRF-3.
