五灵胶囊对实验性肝硬化大鼠的影响

目的 :观察五灵胶囊对大鼠肝纤维化的作用及其机制。方法 :采用以四氯化碳 (CCl4)为主的复合因素致大鼠肝纤维化 ,化学法测定血清羟脯酸 (HYP)含量 ,放免法测定血清与肝匀浆I型前胶原 (PCI)和Ⅲ型前胶原 (PCⅢ )水平 ,免疫组化法检测肝组织肿瘤坏死因子 (TNF)、纤维结合蛋白 (FN )的表达。同时 ,体外培养成纤维细胞 ,观察五灵胶囊对其的抑制作用。结果 :五灵胶囊能显著降低肝纤维化大鼠血清PCI、PCⅢ和肝匀浆HYP、PCI。五灵胶囊组大鼠肝FN表达量明显少于模型组。体外实验表明 ,五灵胶囊可抑制成纤维细胞的生长。结论 :五灵胶囊确有抗肝纤维化作用

Effects of Wuling Capsules on Experimental Hepatic Fibrosis in Rats

AIM： To investigate the effects and mechanism of Wuling capsuleson liver fibrosis in rats METHODS： Hepatic fibrosis models were induced by carbon tetrachloride（ CCl4） in rats The hydroxyproline in the serum and the liver homogenate were detected by chemical method， the precollogen type I（ PC I） and precollogen type Ⅲ （ PCⅢ ） were measured by radioimmunoassay， and the tumor necrosis factor（ TNF） and fibronectin（ FN） were observed by histochemistry method In addition， fibroblasts were cultured in vitro， and the inhibitory effects of Wuling capsules on the cells were also observed RESULTS： Wuling capsules could significantly reduce the levels of PC I and PCⅢ in the serum and the levels of hydroxyproline and PCⅢ in the liver homogenate from rats with hepatic fibrosis The expression of FN in the livers of Wuling capsules treated group was lower than that in the livers of the control group Moreover， in vitro experiment showed that Wuling capsules could inhibit the growth of fibroblasts CONCLUSION： Wuling capsules could inhibit the development of liver fibrosis in rats
