凋亡相关分子在非小细胞性肺癌与癌旁组织中的表达及其意义

摘    要:目的探讨凋亡信号分子CASP3、CASP8、CASP9与抗凋亡信号分子Bcl-2、XIAP在肺癌与癌旁中的表达差异及其意义。方法制备非小细胞肺癌患者的癌组织与癌旁组织的配对石蜡标本的组织芯片。采用免疫组织化学方法检测分析凋亡信号分子CASP3、CASP8、CASP9及抗凋亡信号分子XIAP、Bcl-2在组织中的表达水平。结果配对的癌组织与癌旁组织中凋亡相关分子的表达水平分析,CASP3、CASP9、Bcl-2、XIAP等分子在癌组织表达均有显著升高(P<0.01)。CASP8在癌与癌旁两组间表达水平差异无统计学意义(P>0.05)。双变量相关分析显示癌旁Bcl-2与癌旁CASP8之间(r=0.490,P=0.015)、癌组织中CASP3与CASP9之间(r=0.575,P=0.003)相关性差异均存在统计学意义。结论肺癌组织较癌旁组织凋亡及抗凋亡分子表达均上调,CASP3和Bcl-2分别可能作为潜在的干预靶点而促进肿瘤细胞凋亡。癌旁Bcl-2与癌旁CASP8、癌组织中CASP3与CASP9的表达呈正相关,提示这些分子之间可能存在功能上的相互作用。

Expression of apoptotic signal molecules in NSCLC and adjacent tissues and its significance

Abstract:Objective To investigate the expressions of the pro-apoptotic signal molecules CASP3,CASP8,CASP9 and anti-apoptotic signal molecules Bcl-2 and XIAP in lung cancer and their potential significance.Methods Twenty-six cases of paired cancer versus adjacent non-cancerous tissues,formalin-fixed paraffin-embedded,of NSCLC were collected and made into tissue microarray(TMA) with tissue microarrayer of Beecher Instruments.Immunohistochemistry(IHC) was performed to analyze the expression levels of CASP3,CASP8,CASP9,XIAP and Bcl-2.Results In the evaluable 25 cases of paired cancer versus adjacent tissues,paired sample t test showed that CASP8 was not significantly differentially expressed between cancer and adjacent tissues(P0.05),other molecules CASP3,CASP9,XIAP and Bcl-2 all expressed in cancers at a higher level than those in adjacent tissues(P0.01).Bi-variate correlation analysis primarily found significant correlation between Bcl-2 expression in adjacent tissues and CASP8 in adjacent tissues(r=0.490,P=0.015),and between CASP3 expression in cancers with CASP9 expression in cancers(r=0.575,P=0.003).Conclusions The expressions of the pro-apoptotic signal molecules and anti-apoptotic signal molecules in lung cancer are both up-regulated,CASP3 and Bcl-2 might be of clinical implication as potential target to induce tumor cell apoptosis.Significant correlation between Bcl-2 and CASP8 in adjacent tissues and between CASP3 and CASP9 in cancer tissues suggests that these molecules might have potential functional interaction.
