摘 要: | 目的探讨微波辐射非热效应对人肺癌A549细胞自噬的影响。方法用冰浴法制备微波辐射非热效应模型,剂量为100 W-12 mV/cm2、150 W-18 mV/cm2和200 W-21 mV/cm2,照射10分钟;24小时后,免疫印记法检测细胞自噬相关蛋白Beclin-1、LC3的变化,泛素化蛋白表达变化;间接免疫荧光法检测自噬标志蛋白LC3和p62的变化。结果微波辐射非热效应作用A549细胞,导致自噬相关蛋白Beclin-1表达增加,LC3和p62活化。结论微波辐射非热效应能够诱导人肺癌A549细胞发生自噬。

Abstract: | Objective To investigate the effect of autophagy induced by microwave radiation non-thermal effect on human A549 cell line.Methods Ice bath was employed to establish microwave radiation non-thermal effect model,and dosages were 100 W-12 mV/cm2,150 W-18 mV/cm2 and 200 W-21 mV/cm2 for 10 minutes.After 24 hours,Western blot was used to detect the expression of Beclin-1,LC3 and ubiquitinated proteins.Indirect immunofluorescence was used to dectect the expression and location of the LC3 and p62 proteins.Results The non-thermal effect of microwave radiation on human A549 cells could increase the expression of Beclin-1,and induced activation of LC3 and p62.Conclusion The non-thermal effect of microwave radiation on human A549 cells could induce autophagy. |
