肝细胞癌中Lumican和Parkin的表达及意义

目的探讨肝细胞癌组织中Lumican蛋白及Parkin蛋白的表达及意义,并分析两者的相关性。方法应用免疫组织化学S-P法检测62例肝癌患者癌组织及62例癌旁组织中Lumican及Parkin蛋白的表达并分析其与临床病理因素之间的关系。结果 (1)肝癌组织中Lumican、Parkin的阳性表达率分别为35.5%、32.3%,与癌旁组织的阳性表达率比较(66.1%、72.6%),差异具有统计学意义(P<0.05);(2)Lumican蛋白的表达与Edmondson分级及是否伴有门静脉癌栓相关(P<0.05),而与性别、肿瘤大小、HBsAg、肝硬化和AFP无关(P>0.05);(3)Parkin蛋白的表达与患者肝癌Edmondson分级、AFP水平、是否伴有门静脉癌栓相关(P<0.05),而与性别、肿瘤大小、HBsAg和肝硬化无关(P>0.05);(4)Lumican与Parkin呈正相关(r=0.450;P<0.05)。结论 Lumican与Parkin蛋白在肝细胞癌组织中表达率均较低,可能与肝癌的发生、发展有关,联合检测两者对肝癌的诊断和预后有着重要意义,有望成为肝癌治疗的新靶点。

Expression of Lumican and Parkin in hepatocellular carcinoma and its significance

Objective To investigate the expression and significance of Lumican protein and Parkin protein in hepatocellular carcinoma,and to study their correlation. Methods The expressions of Lumican protein and Parkin protein in 62 cases of HCC and 62 cases of para-carcinoma liver tissues were analyzed by immunohistochemistry method. Results(1)The expression negative rate of Lumican protein(35.5%) and Parkin protein(32.3%) in hepatocellular carcinoma was significantly higher than that in the para-carcinoma liver tissues(P<0.05).(2)Lumican expression had no correlation with gender,tumor size,AFP level,HBsAg and cirrhosis,but had correlation with Edmondson grade and thrombus in the portal vein(P<0.05).(3)Parkin expression had no correlation with gender,tumor size,cirrhosis and HbsAg,but had correlation with Edmondson grade,AFP level,and thrombus in the portal vein(P<0.05).(4)The expression of Lumican protein was positively associated with Parkin(r=0.450,P<0.05). Conclusion Lumican and Parkin may play an important role in the development and progression of liver cancer and may be a new target for cancer therapy.
