MageD1在口腔鳞癌中的表达及其促进凋亡作用的研究

摘    要:目的:检测口腔鳞癌组织和口腔鳞癌细胞系中黑色素瘤相关抗原D1( MageD1)的表达,并在口腔鳞癌细胞系HN6中通过转染慢病毒的方法过表达MageD1,探讨其对细胞凋亡的影响。方法:收集临床标本,通过Western blot和RT?PCR的方法检测口腔鳞癌以及HN6细胞系中MageD1的表达量,用载有MageD1的慢病毒转染HN6细胞,检测转染效果和Bax、Cleaved?Casepase3等凋亡相关蛋白的表达,并采用裸鼠荷瘤实验研究肿瘤体内生长的情况。结果:MageD1在口腔鳞癌组织及HN6细胞株中较正常组织呈低表达,Western blot及RT?PCR结果表明,MageD1成功转染并得到稳定过表达MageD1的Lv?Maged1细胞,并且过表达MageD1的HN6细胞中Cleaved?Casepase3和Bax的表达量高于HN6细胞,荷瘤实验证明MageD1具有抑制肿瘤生长作用( P<0.05)。结论:MageD1在口腔鳞癌组织及细胞系中表达降低,过表达MageD1能促进肿瘤细胞凋亡。

Expression of MageD1 in oral squamous cell carcinoma and its role in promoting apoptosis
Abstract:Objective:To study the expression of MageD1 in oral squamous cell carcinoma ( OSCC) and HN6 cell lines and to ex?plore its role in promoting cell apoptosis through the transfection of MageD1 in the oral squamous cell carcinoma cell line HN6. Meth?ods:The expression of MageD1 in OSCC and HN6 cell lines are tested by Western blot and RT?PCR, and Cleaved?Casepase3 and Bax were tested by Western blot through the transfection of MageD1 in the cell line HN6, and study the growth of cell line HN6 transfected with MageD1 in nude mice. Results:The expression of MageD1 in OSCC and HN6 cell lines are both lower than normal tissues. Ma?geD1, Cleaved?Casepase3 and Bax were upregulated in the Lv?Maged1?HN6 cells than the HN6 cells. The implantation experiment showed that tumors formed by the Lv?Maged1 cells are smaller than those by HN6 cells in nude mice. Conclusions:MageD1 is down?regulated in the OSCC and the over?expression of MageD1 promotes cell apoptosis.
