人参皂苷Rg3对涎腺腺样囊性癌SACC-M细胞的增殖和凋亡的影响

摘    要:目的研究人参皂苷Rg3对涎腺腺样囊性癌SACC-M细胞的增殖和凋亡的影响。方法用终浓度为10μg/m L、50μg/m L、100μg/m L、150μg/m L、200μg/m L的人参皂苷Rg3处理SACC-M 24、48、72 h后,用MTT法、流式细胞仪检测和观察SACC-M细胞的生长抑制率和凋亡情况。Real-time PCR检测凋亡基因bax、caspase-3的表达。EMSA法检测NF-κB活性。结果10~200μg/m L的人参皂苷Rg3均可显著性抑制SACC-M细胞的增殖并诱导其凋亡;且200μg/m L的Rg3诱导的凋亡率最高,为88.69%;Real-time PCR结果表明人参皂苷Rg3可以显著性诱导凋亡基因bax,caspase-3表达水平的上升(P<0.05);EMSA结果表明人参皂苷Rg3可以抑制SACC-M细胞中NF-κB的活性。结论人参皂苷Rg3可能通过抑制NF-κB的活性,从而增加凋亡基因bax、caspase-3的表达,最终抑制SACC-M细胞的增殖并诱导其凋亡。

Effect of Ginsenoside Rg3 on proliferation and apoptosis of human salivary adenoid cystic carcinoma cells
Abstract:Objective To investigate the effect of Ginsenoside Rg3 on proliferation and apoptosis of human salivary adenoid cystic carcinoma cells. Methods SACC?M cells were treated with Ginsenoside Rg3 ( 10μg/mL, 50μg/mL, 100μg/mL, 150μg/mL, 200μg/mL) for 24, 48, 72 h respectively. The inhibition of cell proliferation was detected by MTT assay, and the rate of apoptosis was measured by flow cytometer. The activity of NF?κB was measured by EMSA. Results 10-200μg/mL Ginsenoside Rg3 could signifi?cantly inhibit the proliferation and induced the apoptosis of SACC?M cells. 200μg/mL Ginsenoside Rg3 could induce the highest rate of apoptosis (88.69%). The results of Real?time PCR proved that Ginsenoside Rg3 could induce the expression level of bax and caspae?3. EMSA assay indicated that Ginsenoside Rg3 could suppress the activity of NF?κB. Conclusion Ginsenoside Rg3 could increase the expression level of bax and caspase?3 through inhibiting the activity of NF?κB, thus finally suppressing the proliferation and then indu?cing apoptosis in SACC?M cells.
