摘    要:目的:探讨Ang-1、Ang-2及Tie2 mRNA在骨肿瘤组织中的表达及其意义。方法:采用逆转录聚合酶链反应法对正常骨组织、骨软骨瘤、尤文肉瘤和骨肉瘤组织标本各6例内Ang-1、Ang-2及Tie2 mRNA含量进行半定量检测。结果:恶性骨肿瘤组织(尤文肉瘤、骨肉瘤)内Ang-2 mRNA/Ang-1 mRNA比值较正常骨组织和良性肿瘤骨组织比值上调。尤文肉瘤和骨肉瘤中Tie2 mRNA的表达上调。结论:恶性骨肿瘤组织内Ang-1,Ang-2及Tie2mRNA表达失衡,肿瘤骨组织内可能存有Ang/Tie2的自分泌和旁分泌系统。深入探讨Ang-1/Ang-2/Tie 2在肿瘤骨组织中的分子生物学机制及其临床病理联系,有望使其成为判定骨肿瘤预后的指标,并为骨肿瘤分子治疗提供靶位点。

Abstract:Objective: To investigate the expression of Ang-1,Ang-2 and Tie2 mRNA in bone tumor tissues and to explore its clinical significance.Methods: Semi-quantative RT-PCR was used to analyze the expression of Ang-1,Ang-2 and Tie2 mRNA in tissues harvested from normal bone,osteochondroma,Ewing's sarcoma and osteosarcoma(n=6).Results: Ratio of Ang-2 mRNA/Ang-1 mRNA was increased in malignant lesions(Ewing's sarcoma and osteosarcoma) when compared with benign lesions(normal bone and osteochondroma).Expression of Tie2 mRNA was up-regulated in Ewing'sarcoma and osteosarcoma.Conclusion: The balance of Ang-1,Ang-2 and Tie2 mRNA levels was disturbed in malignant lesions.There may exist a possible autocrine and paracrine pathway of Angs/Tie2 system in bone malignant lesions.Further investigation of Ang1-,Ang-2 and Tie2 and their biomedical significance in bone tumor tissues and clarify their clinical-pathological relation will make them possible prognostic indices and provide molecular therapy target.
