特异性抑制Aqp1基因对K562细胞生长的抑制作用

摘    要:本研究探讨靶向aqp1(aquaporin 1)基因的siRNA对K562细胞生长的抑制作用。设计siRNA,转染K562细胞,在相差显微镜观察细胞形态变化;用MTT法观察aqp1-siRNA对K562细胞活力的影响;使用流式细胞仪、DNA片段化分析K562细胞凋亡;用RT-PCR半定量检测转染前后aqp1基因表达。结果表明:苏木素-伊红染色后,细胞呈现明显的凋亡形态;转染24小时后aqp1-siRNAs对K562细胞的活性有显著抑制作用;aqp1-siRNA转染K562细胞后可见明显的凋亡峰,24、48和72小时的细胞凋亡率持续上升,分别为24.2%、36.1%和42.9%;48小时后aqp1-siRNA组出现明显的DNA凋亡"梯形电泳带";aqp1基因的表达水平在aqp1-siRNA转染后24、48和72小时,分别减少了33%、46%和57%。结论:aqp1-siRNA能够抑制K562细胞的增殖,诱导其凋亡,可作为治疗恶性肿瘤的新靶点。
