川芎嗪对高氧肺损伤的影响

摘    要:目的探讨川芎嗪对新生大鼠高氧肺损伤的影响。方法选用生后2天体重5~10g的Wistar大鼠160只随机分为Ⅰ、3天空气对照组;Ⅱ、7天空气对照组;Ⅲ、3天高氧对照组;Ⅳ、7天高氧对照组;Ⅴ、3天高氧+川芎嗪组;Ⅵ、7天高氧+川芎嗪组。乳鼠制备成高氧肺损伤模型后第2天起腹腔注射川芎嗪(30mg/kg)。模型制备后于第3、7天时予处死。取肺组织固定、切片,行HE染色及TUNEL染色,观察肺组织病理学改变及测定肺泡上皮细胞凋亡指数;部分动物行支气管肺泡灌洗,收集支气管肺泡灌洗液(bronchoalveolarlavagefluid,BALF),监测总蛋白(TP)及丙二醛(MDA)水平,并与高氧对照组、正常对照组、药物干预组的测定值进行比较分析。结果高氧对照组凋亡指数明显高于空气对照组,有显著性差异(P<0.01)。川芎嗪测定值明显低于生理盐水组,有显著性差异(P<0.01)。高氧对照组BALF中总蛋白、MDA水平明显高于空气对照组,有显著性差异(P<0.01)。川芎嗪BALF中总蛋白、MDA水平明显低于生理盐水组,有显著性差异(P<0.01)。结论川芎嗪能减少肺泡上皮细胞凋亡,减轻高氧所致肺损伤BALF中总蛋白、MDA水平,对高氧肺损伤有防治作用。

Abstract:Objective:To study the effects of TMP on hyperoxia-induced lung injury.Methods:160 newborn rats (2-day old, 5-10 g) were divided into 8 groups: I. 3days normal control group; II. 7days normal control group; III. 3days hyperoxia group; IV. 7days hyperoxia group; V. 3days hypemxia TMP group; VI. 7days hyperoxia TMP group..The newborn rats begin to be mteraperitoneally injected with TMP(30 mg/kg) after being established hyperoxia-induced lung injury model for 2days, then the rats were killed after hypemxia for 3days and 7days to exam the lung histophathological changes stained with HE and TUNEL, to detect the apoptosis index of alveolar wall cells. For paris of rats, to take broncho-alveolar lavage fluid to detect total protein and MDA levels, and to compare the conclusions with normal control group ,hypemxia group and drug group.Results:The apoptosis indexes of hypemxia groups were much higher than that in normal control groups(P<0.01). The apoptosis indexes of drug groups were much lower than that in saline groups(P<0.01). The total protein and MDA levels in BALF of hyperoxia groups were much higher than that in normal control groups(P<0.01). The total protein and MDA levels in BALF of drug groups were much lower than that insaline groups(P<0.01).Conclusion:TMP can decrease cerebrocelMar apoptosis in lung tissues with hyperoxia damage. It also can decrease total protein and MDA levels in BALF of neonatal rats lung with hypemxia damage, so TMP has prevention and cure effect on neonatal rats s lung with hyperoxia damage.
