甘肃产银州柴胡挥发性成分的超临界萃取-气相色谱-质谱联用分析

摘    要:目的:对甘肃产银州柴胡的超临界萃取挥发性成分进行分析。方法:采用超临界二氧化碳(CO2)流体萃取法(SFE—CO2)对银州柴胡进行提取,利用气相色谱-质谱联用技术(Gc—Ms)对提取物中的挥发性成分进行分离分析,并计算各成分的相对百分含量。色谱柱:DB-5MS毛细管柱(30m×0.25mm×0.25μm);进样口温度:250℃;程序升温:初始40℃,以3℃/min升至220℃,保持25分钟,以5℃/min升温至250℃,保持30分钟;载气为高纯氦气,流速1.0mL/min;离子源温度:250℃;离子源电离能:70eV。结果:从银州柴胡中共分离鉴定出25个化合物,占挥发油总量的52.2%。结论:该实验结果为了解银州柴胡的化学成分及进一步开发利用提供了依据。

Abstract:Objective: To analyze volatile components from Gansu Yinzhou ChaiHu (Bupleurum yinehowense Shah et Y.Li) obtained by supercritical CO2 fluid extraction(SFE-CO2). Method: Yinzhou ChaiHu was processed with the method of SEF-CO2, volatile components of the extract were analyzed with gas chromatography-mass spectrometry (GC-MS) and relative percentage compositions of the ingredients were calculated. Chromatographic column: DB-5MS column (30 m×0.25 mm×0.25 μm); inlet temperature: 250℃ ; temperature programming: initial 40℃, rising to 220℃ at the rate of 3℃/min, maintaining 25 min, then rising to 250℃ at the rate of 5℃/min, maintaining 30 min; carrier gas was helium, the flow rate: 1.0mL/min; ion source temperature: 250℃; ionization en- ergy: 70eV. Result: There were 25 kinds of compounds identified from Yinzhou ChaiHu, it occupied 52.2% of total contents of volatile oil. Conclusion: The results of the test could provide evidence for further development, the uti- lization and the understanding of chemical ingredients of Yinzhou ChaiHu.
