阿魏酸对活化内皮细胞粘附分子表达的抑制作用

摘    要:目的 观察阿魏酸对活化的人脐静脉内皮细胞表达粘附分子的影响 ,以探讨其抗动脉粥样硬化的部分分子机制。方法 用脂多糖 10mg·L- 1刺激培养的人脐静脉内皮细胞 4h诱导其表达E 选择素、刺激 18h诱导其表达血管细胞粘附分子 1;用过氧化氢 3 0 0 μmol·L- 1刺激 2h诱导其表达P 选择素 ,部分细胞在刺激前 3 0min用阿魏酸 0 2 1、0 41或 0 62mmol·L- 1预处理 ,用流式细胞术检测内皮细胞粘附分子表达水平。结果 阿魏酸抑制内皮细胞E 选择素及P 选择素表达 ,轻度抑制血管细胞粘附分子 1表达 ,同时抑制内皮细胞vWF的释放。结论 阿魏酸可通过抑制内皮细胞粘附分子表达而发挥抗动脉粥样硬化作用

Abstract:AIM To investigate the effects of ferulic aci d (FA) on the expression of adhesion molecules by activated human umbilical vein e ndothelial cells (HUVEC) and to elucidate one of the molecular mechanisms of FA 's anti-atherosclerosis effect. METHODS Cultured HUVEC were tr e ated with ferulic acid (0 41 mmol·L -1 or 0 62 mmol·L -1 ) and stim ulated with lipopolysaccharide (LPS 10 mg·L -1 ) 30 min later. Expression of E-selectin and vascular cell adhesion molecule-1 (VCAM-1) were de- tected b y flow cytometry (FCM) after 4 h and 18 h respectively . Cultured HUVEC were treated with ferulic acid (0 21, 0 41 mmol·L -1 or 0 62 mmol·L -1 respectively) and stimulated with H 2O 2 (300 μmol·L -1 ) 30 min later. After 2 h, expression of P-selectin on HUVEC surface wa s dedected by FCM. RESULTS Ferulic acid inhibited the expression of E-selectin and P-selectin on activated HUVEC. The inhibitive effect of feru lic acid on expression of VCAM-1 was slight. It also suppressed the secrection of vWF on activated HUVEC. CONCLUTION The inhibitive effects of f erulic acid on the expression of adhesion molecules on activated HUVEC may contr ibute to its anti-atherosclerosis effects.
