腰神经根压迫性损伤及药物干预后BDNF和NT-3含量的研究

摘    要:目的:观察大鼠压迫损伤后腰神经根组织BDNF和NT-3的含量;探讨损伤后不同阶段神经根组织神经营养因子的表达,及药物腰痛宁和弥可保对其表达的影响。方法:采用置入微型硅胶片法,建立大鼠腰神经根压迫性损伤模型,用酶联免疫吸附试验(ELISA法),测定损伤后30、60天神经根组织中BDNF和NT-3含量,观察腰痛宁及弥可保等药物干预对其表达变化的影响。结果:正常时,腰神经根组织中BDNF和NT-3有一定量的表达。损伤30天时明显升高(P〈0.01)。60天下降接近正常组。损伤30天,腰痛宁组BDNF、弥可保组BDNF、NT-3的表达明显低于模型组,差异有统计学意义(P〈0.01)。损伤60天,腰痛宁组BDNF、NT-3及弥可保组BDNF的表达均高于模型组,差异有统计学意义(P〈0.05)。结论:腰神经根压迫损伤30天时,损伤神经根组织BDNF、NT-3的表达自身保护性、反应性升高,可能与促进损伤神经再生修复有关。腰痛宁、弥可保均可影响和调控损伤30、60天不同阶段神经根组织BDNF、NT-3的表达,参与损伤神经再生修复过程。

Abstract:Objectives: To observe the effect of Yaotongning (YTN) and Methycobal on BDNF and NT-3 in nerve root compression injury rats. Methods:The nerve root compression injury models were established by inserting mini silicon film to nerve root. EI.ISA was applied to evaluate the change of BDNF and NT-3 in nerve root tissue at the 30th and 60th day. Results:The BDNF and NT-3 content in lumbar nerve root significantly increased after at the 30th day (P〈0.01), and recovered to the normal level at the 60th day. BDNF content inYTN and Methycobal groups and NT-3 in YTN group were markedly lower than those in model group at the 30th day (P〈0.01). And BDNF and NT-3 content of YTN group and NT-3 content in Methycobal group were statistically higher than model group at the 60th day. Conclusion: The increased expression of BDNF and NT-3 in nerve tissue at the 30th day when injury might result in damaged nerve tissue regeneration. YTN and Methycobal can modulate the expression of BDNF and NT-3, which participate in the damaged nerve tissue regeneration process.
