眼针对脑缺血再灌注损伤72h大鼠海马组织ICAM-1表达的影响

摘    要:目的:观察眼针对脑缺血再灌注损伤模型大鼠脑组织细胞间黏附因子1(intercellular adhesionmolecule-1,ICAM-1)表达的影响。方法:64只SD大鼠随机分为:正常组(未处理)、假手术组(同模型组,仅鱼线插入深度1cm)、模型组(应用线栓法复制脑缺血再灌注损伤大鼠模型,鱼线插入深度1.820cm)和眼针组(模型成功后取肝区、上焦区、下焦区、肾区进行眼针治疗20min,每日2次),每组16只。于再灌注后72h进行神经功能缺损评分,采用Western blot、实时荧光定量聚合酶链反应(RQ-PCR)方法测定海马组织ICAM-1蛋白及mRNA表达的变化。结果:眼针治疗后可使大鼠脑梗死灶体积减小,并显著降低海马组织ICAM-1蛋白及mRNA表达(P0.01)。结论:眼针具有明显的改善大鼠脑缺血再灌注损伤的作用,其机制与下调海马组织ICAM-1表达有关。

Abstract:Objective: To investigate the effect of the eye acupuncture therapy on hippocampus ICAM-1 expression in rats with acute cerebral ischemia-reperfusion injury for 72 hours and the related mechanism.Methods: Establish the rat models of cerebral ischemia-reperfusion by suture method.After reperfusion 3h,the neurophysical behaviours were accessed by ZeaLonga neurophysical impairment marks.The methods of Western-blotting and RQ-PCR were taken to detect the alteration of the ICAM-1 protein and ICAM-1 mRNA in rats’ hippocampus after the eye acupuncture therapy.Results: Compared with the model group,the neurologic impairment score of the eye-acupuncture group decreased obviously;The expression of ICAM-1 protein and ICAM mRNA in rats’ hippocampus after the eye acupuncture therapy was strongly decreased(P〈0.01).Conclusion: The eye acupuncture therapy can improve the cerebral ischemia-reperfusion injury evidently by decreasing the expression of hippocampus ICAM-1 protein.
