摘    要:目的:研究LY294002对Raji细胞化疗的增敏作用。方法:Westernblot法测定LY294002处理后,Raji细胞pAKt蛋白变化。用LY294002、阿霉素、两药联合及序贯处理细胞,POD法测定各组细胞凋亡率。结果:LY294002可显著降低Raji细胞中pAKt蛋白的表达水平;序贯处理组细胞凋亡率比联合用药及其它两组明显提高。结论:抑制P13K/Akt信号转导通路可提高淋巴瘤化疗的效果,LY294002可能成为淋巴瘤治疗中潜在的化疗增敏分子靶向药物。

Study on LY294002 for chemotherapeutic sensitization in Raji cell
Abstract:Objective:To investigate the effects on chemotherapeutic sensitization of the LY294002 in Raji cell. Methods Western bloting was used to detect the level of pAKt protein after the Raji cell was treated with LY294002. The Raji ceU was treated with LY294002, or doxorubiein alone or combined or sequentially respectlvely.POD was used to detect the ratio of apeptosis.Resttlls LY294002 decreased the level of phospho- AkT efficiently in the Raji cell line. Sequential use of LY294002 and doxorubicin increased the ratio of apoptosis and significant difference between the combined group and the other two groups. Conclusion Inhibiting the PI3K/Akt signaling pathway can increase the chemotherapeutic sensitivity of lyra- phoma. LY294002 may be a potential molecular therapeutic agent at PI3K pathway in lymphoma.
