普胃丸对慢性胃溃疡大鼠胃组织中EGF和EGFR蛋白表达的影响

摘    要:目的:考察普胃丸(Puwei Pill,PWP)对慢性胃溃疡(chronic gastric ulcer,CGU)大鼠胃组织中促增生因子表皮生长因子(Epidermal Growth Factor,EGF)和表皮生长因子受体(epidermal growth factor receptor,EGFR)蛋白表达的影响。方法:将大鼠随机分为5组,即正常组、假手术组、模型组,PWP组和奥美拉唑肠浴片(OMLZ)组,用醋酸烧灼法建立大鼠CGU模型,通过灌注给药,于16 d后处死大鼠,然后分别采用免疫组化法和Western blot法,观察PWP对胃组织中EGF和EGFR蛋白表达的影响,考察PWP抗CGU的促黏膜增生机制。结果:与模型组比较,PWP与OMLZ组EGF和EGFR的平均阳性密度均显著增加(P<0.01或P<0.05),EGF和EGFR蛋白表达均显著提高(P<0.01或P<0.05),PWP可明显增加胃组织中EGF和EGFR的蛋白表达。结论:PWP通过促进胃组织中EGF和EGFR的蛋白表达来发挥抗CGU的作用。

Abstract:Objective: To investigate the effects of Puwei Pill (PWP) on expressions of epidermal growth factor (EGF) and epidermal growth factor receptor (EGFR) protein in gastric tissue in rats with chronic gastric ulcer(CGU). Method: The animals were divided into five groups. CGU model was built by using acetic acid. Then the protein expressions of EGF and EGFR in gastric tissue was determined by immunohistochemical staining and western blot so as to evaluate the pro-mucoca growth mechanism underlying PWP resisting of CGU. Result: Compared with the model,PWP could obviously increase the expressions of EGF and EGFR protein in gastric tissue. Conclusion: PWP plays the role of resisting of CGU through increasing the expression of EGF and EGFR protein in gastric tissue.
