刺五加总苷及刺五加苷B对持续黑暗条件下果蝇睡眠-觉醒节律的量效关系研究

摘    要:目的:探讨刺五加总苷及苷B改善睡眠-觉醒节律的量效关系。方法:以果蝇为实验对象,在持续黑暗条件下,总苷固定给药7 d,苷B给药3 d,采用DAMS监测果蝇活动及睡眠状态,分析比较其平均睡眠时间、睡眠-觉醒节律和睡眠时间百分比。结果:刺五加总苷对于果蝇睡眠时间表现低剂量增加(P<0.05)高剂量减少(P<0.05),增加片段化睡眠,略微增多白天睡眠时间百分比。刺五加苷B可通过增加果蝇夜晚睡眠时间(P<0.001)进而延长全天睡眠时间(P<0.01),增加片段化睡眠,增多白天及夜晚睡眠时间百分比。结论:刺五加总苷、苷B均能改善果蝇睡眠,其最佳剂量为0.125%(即生药量7.125 mg·kg^-1)总苷、1%苷B。

Study on the dose-effect relationship of acanthopanax total glycoside and eleutheroside B on sleep-wake rhythm in drosophila melanogaster under continuous darkness

Abstract:Objective:This study aims to investigate the dose-effect relationship of acanthopanax senticosus and eleutheroside B in improving sleep-wake rhythm.Methods:Drosophila melanogaster was administered with acanthopanax senticosus for 7 days and eleutheroside B for 3 days under the condition of darkness,and the activity and sleep state were monitored by DAMS.After that,the sleep time,sleep-wake rhythm and percentage of sleep time were analyzed and compared.Results:Decreased acanthopanax senticosus led to more sleep time of drosophila melanogaster(P<0.05),and increased dose resulted in less sleep time(P<0.05).This also increased the fragmented sleep and slightly increased the percentage of daytime sleep time.Eleutheroside B could prolong the entire sleep time(P<0.001)by increasing the night sleep time(P<0.001),so as to increase the fragmented sleep time and the percentage of the day and night sleep time.Conclusion:Acanthopanax senticosus and eleutheroside B can improve the sleep of drosophila melanogaster.The optimum dosage is 0.125%(dried medicinal herbs is 7.125 mg·kg^-1)of acanthopanax senticosus and 1%of eleutheroside B.
