精子获能前后运动轨迹变化与MTATP_8、MTCYB基因缺失

摘    要:目的:为探讨精子MTATP8 、TMCYB基因缺失与精子获能障碍导致男性不育的关系。 方法:60 例男性不育者(其中30 例精子活动力正常,30 例精子活动力异常)和30 例生育男性精子获能前后运动轨迹变化,以及用PCR技术检测其线粒体脱氧核糖核酸(m tDNA)基因缺失情况。 结果:男性不育组获能后轨迹没有变化的共检出16 例,其中活动力异常组检出10例,活动力正常组检出6 例;生育组获能前后运动轨迹均有变化。不育组活动力异常的30 例标本中检出MTATP8 基因缺失2 例,检出MTCYB基因缺失4 例;不育组活动力正常的30 例标本中检出MTATP8 基因缺失2 例,检出MTCYB基因缺失3 例,经统计两组差异不显著(P> 0.05)。生育组30 例标本未检出MTATP8、MTCYB基因缺失,与男性不育组差异显著(P< 0.05)。不育组MTATP8 基因、MTCYB基因缺失率分别为7% 、12% 。 结论:MTATP8 、MTCYB基因缺失影响精子获能,可能导致男性不育。

Association of MTATP_8,MTCYB deletions with the changes of motive tracts before and after sperm capacitation

Abstract:Objectives:To explore the relationship between deletions of MTATP 8,MTCYB gene and sperm capacitation defection leading to mlae infertility. Methods:The deletion of MTATP 8 and MTCYB genes before and after sperm capacitation in 60 infertile (30 with poor sperm motility and 30 with normal sperm motility) and 30 fertile males were analyzed by PCR.The changes of motive tracts of sperm were observed simultaneously. Results:Among 60 samples from infertile males,16(10 with poor sperm motility and 6 with normal motility) were found to have no change on motive tracts before and after sperm capacitation.All the 30 samples from fertile males were found to have great changes on motive tracts before and after sperm capacitation.In the 60 samples from infertile males,2 MTATP 8 deletion and 4 MTCYB deletion were detected in the 30 samples with normal motility,while 2 MTATP 8 deletion and 3 MTCYB deletion were detected in the 30 samples with poor motility.No significant difference was found between the 2 groups( P >0.05).The deletion of MTATP 8 gene or MTCYB gene was not found in the 30 samples from fertile males.The difference of MTATP 8 gene or MTCYB gene deletion between the infertile and fertile group were significant( P <0.05).In infertile males,the rate of MTATP 8 gene deletion was 7%,while the rate of MTCYB gene deletion was 12%. Conclusions:The deletion of MTATP 8 gene or MTCYB gene may affect sperm capacitation,which may lead to infertility.
