| 摘 要： | 目的:探讨在小鼠受精卵中,PKC及PKB是如何通过p21CIP1/WAF1蛋白影响小鼠受精卵G2/M的转换。方法:以小鼠受精卵为材料,通过Westernblot及免疫荧光的方法,检测经PKC及PKB抑制剂处理后的p21蛋白表达及定位。结果:经PKC抑制剂(PMA)处理后的小鼠G2期受精卵中p21蛋白表达增加,而经PKB抑制剂(LY294002)处理后的p21蛋白表达没有变化,但是其在亚细胞的定位发生了改变。结论:PKC通过改变p21蛋白表达,影响小鼠受精卵G2/M转换。而PKB通过改变p21蛋白在小鼠受精卵中的亚细胞定位,影响受精卵G2/M转换。

| Abstract: | Objective: To explore effect of PKC and PKB on G2/M Transition by Affecting p21 in mousefertilized eggs.Methods: The expression and subcellular localization of p21 in fertilized eggs were detected byWestern blotting and immunocytochemistry after treated with PKB and PKC inhibitors.Results: PKC inhibitorinduce upregulation of p21 protein in mouse fertilized eggs in G2 phase,while PKB had no effect on it.p21 proteinin G2 phase was located in the cytoplasm and nucleus,redistributing in the nucleus treated with PKB inhibitor.Conclusion: The PKC controlled G2/M transition by affecting the expression of p21 protein while PKB controlledG2/M transition by changing the subcellular localization of p21 protein. |
