摘    要:应用自制电致细胞融合仪,在显微镜直视下,可观察到小鼠胸腺细咆时伊红染料的电渗入效应、在电极间各类细胞的电介质电泳现象以及植物原生质体间的电融合过程.小鼠脾细胞和骨髓瘤FO细胞电融合后,转移至HAT培养基继续培养后,有细胞克隆形成,此系电融合致杂交瘤形成的可靠依据.以上结果表明,自制电融合仪能满足细胞工程研完的基本要求.

Abstract:A series of cytobiologieal experiments was carried out with a self-made electrofusion apparatus XR-1. The electropermeabilization of eosine dye by mouse thymocytes, the dielectrophoresis of various types of cells, electrofu-sion between protoplasts of the domesticated tomato (Lycopersicon esculen-tum)and the wild tomato (L.hirsutum) were directly monifored under the inverse microscope. Seven days after the fusion of mouse spleen cells and mouse myeloma FO cells, the hybridoma cell clones were formed in HAT mediam which was a strong evidence of the success in electrofusion bet-ween these two types of cells. The above rasults indicate that the self-made electrofusion apparatus is able to meet the basic requirements of electro-fusion and electropermeabilization.
