同型半胱氨酸体内外诱导鼠胚胎心肌细胞凋亡的研究

摘    要:目的 探讨同型半胱氨酸(HCY)是否能诱导啮齿类动物胚胎心肌细胞凋亡及剂量与凋亡的关系。 方法 应用大鼠胚胎心肌细胞体外培养及小鼠体内致畸实验,加入4-20 mmol/L的HCY,经荧光显微镜及透射电镜观察细胞凋亡情况。 结果 体外心肌细胞培养中,心肌细胞凋亡的比例随着HCY剂量的增加而增加。培养12 h出现凋亡,14 h达到高峰。小鼠体内致畸实验,给予孕鼠HCY 0.74 mmol/kg,胚胎心肌和脑组织中凋亡细胞明显可见。 结论HCY可以诱导胚胎心肌细胞凋亡,可能是HCY导致胚胎发育异常的机制之一。

Abstract:Objective To explore the possible effect of homocysteine (HCY) in inducing apoptosis in murine embryonic ventricular myocytes and the dose-response relationship. Methods In vivo mouse teratology test and in vitro rat embryonic ventricular myocyte culture were used. Cell apoptosis was observed at different time point by fluorescence microscopy or electron microscope. Results The percentage of cell apoptosis increased with the increase of HCY concentration, and reached its maximum at 14 h of culture. In vivo test showed apparent apoptosis of heart and brain cells when HCY was administered at the concentration of 0. 74 mmol/kg or higher. Conclusion HCY can induce ventricular myocyte and brain cell apoptosis in embryos, which may contribute to the dysmorphogenesis of embryos.
