低温及蟾酥对麻醉家兔心肌缺血再灌注损伤的保护作用研究

摘    要:目的在家兔心肌缺血再灌注模型上观察低温和蟾酥及两者合用对心肌缺血再灌注损伤的保护作用.方法将30只新西兰大耳白兔随机分为对照组(A组)、缺血再灌注组(B组)、物理降温组(C组)、蟾酥组(D组)和物理降温 蟾酥组(E组),并分设基础、结扎前30 min、结扎或穿线即刻、结扎或穿线后30 min和旷置或再灌注30 min、60 min、90 min、120 min8个时点,动态监测体温,监测再灌注120 min时血液及缺血区中央心肌组织三磷酸腺苷(ATP)、超氧化物歧化酶(SOD)、丙二醛(MDA)、游离脂肪酸(FFA)和乳酸(LD)的含量.结果心肌缺血再灌注损伤120 min的家兔与对照组比较,干预组与缺血再灌注比较,明显改善血清及心肌MDA、FFA、LD和SOD含量,显著升高心肌ATP活性(P<0.05),同时还观察到联合应用降温和蟾酥的效果优于单独应用降温组或蟾酥组(P<0.05).结论低温及蟾酥对家兔心肌缺血再灌注损伤具有明显保护作用,两者合用保护作用更显著.

Abstract:Objective To investigate the protective effects of hopothermia and toadfish on myocardial ischemia/reperfusion injury.Methods Thirty New Zealand rabbits were randomly divided into 5 groups with 6rabbits in every group: A control group;B myocardial ischemia reperfusion group;C hypothermia group;D toadfish group;E hypothermia+toadfish group.Temperature is recorded before,during and at the end of myocardial ischemia and at 30,60,90,120 min of reperfusion.Blood samples are taken for content of plasma SOD,MDA,FFA and LD at the end of reperfusion.At the end of the experiment myocardial tissue 200 mg is obtained from ischemia area for content of myocardial ATP,SOD,MDA,FFAand LD activities.Results Compared with control group,the therapeutic group markedly improved plasma and ischemic myocardial SOD,MDA,FFA and LD and significantly increased ischemic myocardial ATP activity(P<0.05 )vs B group.The group hypothermia and toadfish has more significant effect than topical treatment with hypothermia or toadfish(P<0.05).Conclusion Both hypothermia and toadfish have protective effects on myocardial ischemia/reperfusion injure and combination of them provides better protection.
