摘    要: 目的 初步观察重组腺病毒rAd-MK-TNFα对乳腺癌细胞的抑制作用。方法 体外转染系列重组腺病毒,观察rAd-MK-TNFα在乳腺癌细胞Bcap37中TNFα的表达以及其对乳腺癌细胞的生长抑制作用,并比较其与对照病毒对乳腺癌细胞及人成纤维细胞生长的抑制作用的不同。结果 rAd-MK-TNFα感染Bcap37细胞后,可检测到TNFα的表达;rAd-MK-TNFα对Bcap37乳腺癌细胞存在生长抑制作用,且与其病毒浓度呈正相关;rAd-MK-TNFα对人成纤维细胞的抑制作用小于非靶向腺病毒(rAd-CMV-TNFα)。结论rAd-MK-TNFα可能通过靶向表达TNFα对乳腺癌细胞产生抑制细胞生长的作用。

Abstract:Objective To evaluate the inhibitory effect of recombinant adenovirus rAd-MK-TNFα on cell growth of breast cancer in vitro. Methods The Bcap37 breast cancer cells and human fibroblast cells were infected by rAd-MK-TNFα and control recombinant adenovirus rAd-CMV-TNFα, then the inhibitory effects of both viruses on cell growth were observed and compared. Results TNFα was detected in Bcap37 breast cell after infected by rAd-MK-TNFα, and obvious growth inhibition was observed which was correlated with virus concentration. The inhibitory effect on cell growth of rAd-MK-TNFα on human fibroblast cell was less than that of the rAd-CMV-TNFα. Conclusion rAd-MK-TNF may have tumor cell selective inhibition effect on breast cancer by expressing midkine driven TNFα.
