雌激素对大鼠骨性关节炎影响的实验研究

目的　了解雌激素水平改变对雌性大鼠骨性关节炎的影响。　方法　 SD大鼠 4 0只 ,采用膝关节制动制作骨性关节炎及同时切除卵巢的方法制作绝经期大鼠模型。实验分为 4组 , 组为正常对照组 ; 组为单纯关节固定组 ; 组为关节固定加卵巢切除组 ; 组在 组基础上同时给予 0 .80 mg/ (kg· d)己烯雌酚 ,连续 4周。 1个月后进行骨性关节炎病理学观察、评分和血清骨钙素检测。　结果　 组 (5 .72± 0 .6 4 )μg/ L血清骨钙素明显高于 组 (3.5 0±0 .39)μg/ L及 组 (3.95± 0 .4 4 )μg/ L。骨性关节炎、关节软骨和滑膜组织病理学评分 组 (10 .83± 4 .35、4 .2 1± 2 .0 3) , 组 (15 .32± 3.4 2、7.6 2± 3.4 2 ) , 组 (12 .6 5± 2 .73、5 .4 6± 1.2 3)。　结论　雌性大鼠切除卵巢可加重骨性关节炎的病理改变 ,而应用己烯雌酚替代后可延缓骨性关节炎的病理过程。雌激素可能在延缓骨性关节炎的发病过程中发挥重要作用。

AN EXPERIMENTAL STUDY ON EFFECT OF ESTROGEN ON OSTEOARTHRITIS IN FEMALE RATS

OBJECTIVE: To investigate the effect of estrogen on osteoarthritis in female rats. METHODS: Forty female rats were divided into four groups. In group I, the rats were not given any treatment as a control. In groups II, III and IV, the rats received fixing left knee joint on extension position. Meanwhile, the rats received ovariectomy in group III; ovariectomy and diethylstilbestrol treatment in group IV, respectively. After 4 weeks, histological observation and serum BGP examination were done. RESULTS: In groups II, III and IV, the levels of serum BGP were 3.50 +/- 0.39, 5.72 +/- 0.64 and 3.95 +/- 0.44, respectively. The pathologic grades of cartilage and synovium were 10.83 +/- 4.35 and 4.21 +/- 2.03; 15.32 +/- 3.42 and 7.62 +/- 3.42; and 12.65 +/- 2.73 and 5.46 +/- 1.23, respectively. CONCLUSION: Estrogen may play an important role in delaying the development of osteoarthritis.
