大肠癌p16、cyclin D_1表达及其临床病理意义

摘    要:采用免疫组化中的 S- P法检测 6 7例大肠癌中 p16、cyclin D1 的表达。结果大肠癌 p16、cyclin D1 的阳性表达率分别为 47.8%和 44 .8% ,高分化癌 p16阳性率明显高于低分化癌 (P<0 .0 5 ) ,而低分化癌 cyclin D1 过表达率高于高分化癌 (P<0 .0 5 ) ;淋巴结转移阳性组的 p16阳性率低于淋巴结转移阴性组 ,而淋巴结转移阳性组 cyclin D1 过表达率高于淋巴结转移阴性组 (P<0 .0 5 )。相关分析表明 ,在大肠癌中 p16与 cyclin D1 呈负相关。提示 p16基因失表达和cyclin D1 过表达与大肠癌的分化程度和淋巴结转移有关 ,其在大肠癌中的反向表达支持两者之间可能存在相互抑制机制。

Abstract:To study p16 protein and cyclin D-1 expressed in colorectal carcinoma and their clinical pathological significance , the expressions of the two proteins were detected by using S P immunohistochemical technique in 67 cases of colorectal carcinoma. The positive expression rate of p16 protein and cyclin D-1 were 47.8% and 44.8% respectively. The positive rate of p16 protein in well differentiated carcinoma was higher than that in poorly differentiated carcinoma (P<0.05), while the over expression rate of cyclin D-1 in poorly differentiated carcinoma was higher than that in well differentiated carcinoma (P<0.05). The positive rate of p16 protein with lymph node metastasis was lower than that without metastasis. The over expression rate of cyclin D-1 without lymph node metastasis was lower than that with metastasis. There was a remarkable difference (P<0.05), with an interesting finding that inverse expression between p16 protein and cyclinD-1 was shown in most of the colorectal carcinoma. The result shows that the under expression of p16 and over expression of cyclinD-1 were correlated with the degree of differentiation and lymph node metastasis, and the inverse expression between p16 protein and cyclinD-1 suggest that there is an inhibitory mechanism between these two genes.
