通心络对糖尿病小鼠视网膜组织血管内皮生长因子及色素上皮衍生因子表达的影响

摘    要:目的 观察通心络胶囊对自发糖尿病KK/Upj-Ay小鼠视网膜组织血管内皮生长因子(vascular endothelial growth factor,VEGF)和色素上皮衍生因子(pigment epitheliumderived factor,PEDF)基因及蛋白表达的影响.方法 KK/UN-Ay小鼠40只随机分为模型组、通心络低(1 g/kg)、中(2 g/kg)、高(4 g/kg)剂量4组,通心络灌胃给药12周.以C57BL/6小鼠为对照组.应用实时荧光定量聚合酶链反应(qPCR)法检测各组小鼠视网膜组织中VEGF和PEDF的基因表达;蛋白印迹(western blot)法检测视网膜组织中VEGF和PEDF蛋白的表达.结果 与对照组比较,模型组VEGF mRNA、蛋白表达明显增高(P<0.01),PEDF mRNA、蛋白则明显下降(P<0.01);通心络治疗12周后,与模型组比较,低、中、高剂量组VEGF mRNA和蛋白表达水平均明显下降(P<0.01),而PEDFmRNA和蛋白表达水平明显增高(P<0.01).结论 通心络胶囊可通过抑制自发2型糖尿病小鼠视网膜组织VEGF的表达及提高PEDF的表达来改善视网膜病变.

Abstract:Objective To explore the effect of Tongxinluo capsule on the gene and protein expression of vascular endothelial growth factor(VEOF) and pigment epithelium-derived factor(PEDF) in.the retinal tissue of KK/Upj-Ay mice. Methods Forty KK/Upj-Ay mice were randomly divided into model control group ( n = 10) and Tongxinluo treatment group ( n =30), which were subdivided into three subgroups: low-dose(1 g/kg), median-dose(2 g/kg) and high-dose(4 g/kg) groups. Mice in the tongxinluo treatment group were intragastrically given Tongxinluo capsule for 12 weeks. C57BL/6 mice were used as control group. The mRNA expression of VEGF and PEDF in retina tissue were determined using real time PCR, and their protein expression was determined using Western-blot. Results The mRNA and protein expression of VEGF in retinal tissue of the model control group increased significantly (6.40±0.56 vs 1.26± 0.19 and 0.62 ±0.09 vs 0.16 ± 0.02, respectively, P 〈0.01), while the expression of PEDF decreased remarkably both in mRNA (0.29±0.06 vs 0. 98± 0. 08 , P 〈0.01) and protein (0. 48±0.10 vs 0. 84±0. 08, P 0.01), as compared with those in the control group. After treatment with Tongxinluo for 12 weeks, both the mRNA and protein expression of VEGF were prominently reduced in all three dose groups, while the mRNA and protein expression of PEDF were markedly induced, as compared with the expression of VEGF and PEDF in the model control group ( P 〈0.01). Conclusion Tongxinluo capsule may improve diabetic retinopathy by inhibiting the expression of VEGF and inducing the expression of PEDF in retinal tissue of KK/Upj-Ay mice.
