明胶酶及其抑制剂在慢性格林-巴利综合征中的表达

摘    要:目的 观察MMP-9、MMP-2、TIMP—1、TIMP-2在慢性格林-巴利综合征(CIDP)腓肠神经中的表达部位及水平,探讨它们在CIDP发病机制中的作用。方法 收集CIDP和NIN腓肠神经活检蜡块,用免疫组化分别检测CIDP组和NIN组腓肠神经中MMP-2、MMP-9、TIMP-1和TIMP-2蛋白的表达,光镜观察表达部位,并比较各指标在两组中表达水平的差异性。结果 MMP-9、TIMP-1、TIMP-2在CIDP腓肠神经中的血管、T淋巴细胞和神经纤维均有阳性表达且高于对照组(均P〈0.05)。结论MMP-9在CIDP的发病机制中起重要作用;TIMP-1和TIMP- 2在CIDP中可能通过增加表达来拮抗明胶酶而起保护作用。

Abstract:Objective To examine the expression of MMP-2, MMP-9, TIMP-1 and TIMP-2 in the sural nerve and explore the role of them in the pathogenesis of chronic inflammatory demyelinating polyneuropathy. Methods We detected the localization of MMP-2,MMP-9,TIMP-1 and TIMP-2 in the sural nerve from the patients (20 cases) with CIDP (chronic inflammatory demyelinating polyneuropathy) and the patients (10 cases) with NIN(non-inflammatory neuropathies) using immunohistochemistry and compared the expressing level of them between CIDP and NIN . Results Immunohistochemical findings of the twenty sural nerve biopsies of patients with CIDP disclosed an immunoreactivity of MMP-9,TIMP-1 and TIMP-2 on nerve fibers, endothelial cells and T lymphocytes ,and the expressing levels were higher in the sural nerve of CIDP compared to NIN . Conclusion Our findings strongly suggest that MMP-9 plays an important role in the pathogenesis of CIDP, and increased expressions of TIMP-1 and TIMP-2 in CIDP have a possible role in inhibiting the activity of MMP-9 and MMP-2.
